以大别山地区野生蕙兰(Cymbidium faberi)的花芽和花茎为外植体进行组织培养,考察了1g/LHgCl2消毒时间对外植体生长的影响,并通过正交试验优化诱导愈伤组织的激素浓度组合.结果表明,外植体用1 g/L HgCl2消毒8 min,接种成活率最高;激素浓度组合2 mg/L 2,4-D+4.5 mg/L 6-BA+6 mg/LNAA培养基最有利于外植体的增重;2mg/L 2,4-D+4.0 mg/L 6-BA+6 mg/L NAA有利于诱导花茎和花芽外植体愈伤组织的诱导.%Flower buds and flower stems of Cymbidium faberi were used as explants for callus induction. The effect of 1 g/L HgCl2 disinfecting time on explants was studied. And the combination of plant hormones in medium was optimized by orthogonal design. The results showed that the explants had the highest survive rate when disinfected in 1 g/L HgCl2 for 8 min. The best plant hormone combination for weight increase of explants was 2.0 mg/L 2, 4-D+4.5 mg/L 6-BA+6 mg/L NAA, for callus induction of both flower buds and flower stems explants was 2.0 mg/L 2, 4-D+4.0 mg/L 6-BA+6 mg/L NAA.
