外源性磷酸肌酸r心肌保护多靶作用机制的研究进展

     

摘要

磷酸肌酸(phosphocreatine,PCr)为一种天然高能磷酸化合物,在维护机体能量自稳态中发挥关键作用.外源性PCr已作为心肌保护药广泛用于心血管疾病的治疗中.其结构独特,蕴藏较多生物学信息,为一多靶指向药物.21世纪以来,由于分子生物学的迅速发展,其多靶作用机制不断得到阐明,除能量相关机制外,还包括非能量相关机制,不仅涉及胞内机制,而且涉及胞外机制.该文根据笔者多年研究以及查阅国内外文献,对其多靶作用机制研究进展,包括能量提供、膜稳定化、抗血小板聚集、电生理学、酶抑制以及线粒体保护与抗凋亡作用等方面进行综述.%Phosphocreatine ( PCr), a natural high energy phosphate, plays a pivotal role in maintaining energy homeostasis of the body. Exogenous PCr has been developed as a cardio-protective drug and extensively used in treatment of cardiovascular diseases.PCr has special chemical structure,which confers much bioinformation on it,and becomes a multitarget-directed drug.Since the 21st century,with rapid development of molecular biology,the multiple target action mechanisms of PCr have continually gained elucidation,including energy-related and non-energy-related mechanisms,intracellular and extracellular mechanisms,which are leading to its extensive clinical applications in cardiovascular diseases.Based on author' s research and published literatures,this article reviews the research progress in multiple target action mechanisms of PCr,including energy supply,membrane stabilization, anti-platelet aggregation, electrophysiology, enzyme inhibition and protection of mitochondria, antiapoptotic effect,etc.
