实时荧光定量PCR 法在检测血浆结核杆菌DNA 的应用

         

摘要

Objective:To investigate the application of real-time PCR in plasma dedtcting of mycobac-terium tuberculosis DNA. Method:392 cases of untreated tuberculosis patients were selected in our hospital as experimental group, 100 healthy people were selected as control group. Taking venous blood of two groups, plasma mycobacterium tuberculosis DNA with real-time PCR for detection, and the experimental group with sputum smear examination. Result:The control group with real-time PCR detection were all negative, 80 cases in the experimental group were negative,all sputum smear examination, patients with realtime PCR shows positive ,The negative patients shows 24 cases were positive with real-time PCR. 12 patients became negative after anti-tuberculosis treatment,The experimental group MTB DNA was 1.76 xlO9 copies/ Ml. Conclusion:It shows that TB patients' plasma has MTB DNA, real-time PCR method can be effective for in diagnosing sputum smear-negative patients or sputum without patients.%目的:探讨实时荧光定量PCR 法在检测血浆结核杆菌DNA 的应用及价值.方法:以我院确诊为结核病且未经治疗的患者392 例作为实验组,同时选取健康体检人群100 例作为对照组.分别对两组患者取静脉血,并对其血浆中结核分枝杆菌DNA 采用实时荧光定量PCR 法进行检测,同时对实验组患者进行痰涂片检查.结果:对照组患者实时荧光定量PCR 法检测全为阴性;实验组患者采用痰涂片检查,结果显示有80 例为阴性,其实时荧光定量PCR 法检测结果显示,痰涂片阳性的患者全为阳性,其阴性患者中有24 例为阳性,在对阳性患者抗痨治疗后有12 例患者转为阴性.实验组患者MTB DNA 为1.76×109 copies/mL.结论:本实验表明结核患者血浆中存在MTB DNA,采用实时荧光定量PCR 法可有效的对痰涂片阴性以及无痰患者进行诊断,有较好的利益价值.
