姜黄素激活人膀胱癌 T24细胞自噬诱导细胞凋亡

         

摘要

目的:探讨姜黄素( Cur)对人膀胱癌T24细胞株的抗肿瘤作用及其对细胞自噬的影响。方法采用CCK-8法检测Cur对T24细胞增殖的抑制作用;Annexin V/PI法检测细胞凋亡情况;采用Western blot对细胞LC3B及P62蛋白表达量进行检测;吖啶橙染色观察细胞内自噬囊泡的改变。结果 Cur可以显著抑制T24细胞增殖,诱导细胞凋亡( P <0怂.01)。 Cur诱导T24细胞发生自噬,显著上调LC3蛋白表达,下调P62蛋白表达,增加细胞内自噬囊泡的数量(P<0.01)。 Cur增强顺铂(cis-Dichlorodiamineplatinum,CDDP)的促凋亡作用( P <0.01)。结论 Cur可显著抑制膀胱癌T24细胞的增殖并通过激活细胞自噬促进肿瘤细胞凋亡,Cur可增强CDDP的抗肿瘤效果。%Objective To investigate the anti-tumor effects of curcumin on human bladder cancer cell line (T24) and its effects on cell autophagy in vitro.Methods The inhibition effects of curcumin on cell proliferation of T24 cell line were detected by CCK-8 assay.The cell apoptosis rate was determined by Annexin V/PI assay.The expression levels of LC3B and P62 protein were detected by Western Blot.The changes of autophagic vacuoles were observed by acridine orange staining. Results Curcumin could obviously inhibit the proliferation of T24 cells and induce cell apoptosis ( P <0.01).Curcumin induced autophagy of T24 cells, up-regulated expression levels of LC3 protein, down-regulated expression levels of P62 protein and increased the numbers of autophagic vacuoles ( P <0.01 ) .Moreover curcumin enhanced the apoptosis effect of cis-Dichlorodiamineplatinum (CDDP, P <0.01).Conclusion Curcumin can obviously inhibit the proliferation of T24 cells and induce cell apoptosis through cell autophagy, moreover, curcumin can enhance the anti-tumor effects of CDDP.
