非小细胞肺癌PKM2的表达及与放疗敏感性的关系分析

摘要

Objective To investigate the expression of PKM 2 in non-small cell lung cancer and its correlation with radiotherapy sensitivity .Methods Sixty-eight patients with non -small cell lung cancer who were treated in our hospital from July 2013 to July 2014 were enrolled in the study .All the patients were diagnosed by clinical and operative pathology and were treated by radiotherapy .According to radiotherapy sensitivity , these patients were divided into group A ( sensitivity to radiotherapy ) and group B ( non -sensitivity to radiotherapy ) .The expression levels of PKM 2 in surgical ablation tumor tissues before radiotherapy were detected by immunohistochemistry , the expression levels of PKM 2 mRNA were detected by RT-PCR and the expression levels of PKM 2 protein were detected by Western Blot .Results The immunohistochemistry examination showed that the high expression rate of PKM2 in group A and group B was 21.62%(8/37)and 58.06%(18/31), respectively,there was a significant difference between two groups ( P <0.05).The results by RT-PCR showed that the expression levels of PKM2 mRNA in group B were significantly increased ,as compared with those in group A ( P <0.05). Western Blotting revealed that the expression levels of PKM 2 protein in group A were obviously decreased ,as compared with those in group A ( P <0.01).Conclusion The expression of PKM2 is negatively correlated with radiotherapy sensitivity , which shows that the expression of PKM 2 can be regarded as an important molecular mark to predict radiotherapy sensitivity in patients with non-small cell lung cance so as to realize individualized treatment .%目的 探讨非小细胞肺癌患者PKM2的表达情况与放疗敏感性的关系.方法 取门诊就诊的非小细胞肺癌患者68例为研究对象,所有患者病例均经临床和手术病理确诊.对入组患者放射治疗,根据放疗敏感性分为A、B 2组:A组为对放疗敏感患者,B组为对患者不敏感患者.取放疗前手术切除的肿瘤组织,免疫组化技术法检测PKM2蛋白的表达情况;RT-PCR法检测PKM2的mRNA表达情况;Western blot检测PKM2蛋白表达情况.结果 通过免疫组化发现,PKM2蛋白在A组和B组的高表达率分别为21殮.62%(8/37)和58.06%(18/31),2组比较差异有统计学意义(P<0.05).RT-PCR发现,与A组比较,B组PKM2 mRNA的表达明显上升(P<0.05).Western blot检测结果显示,与B组比较,A组PKM2蛋白表达水平明显下降,差异有统计学意义(P<0.01).结论 PKM2的表达与非小细胞肺癌的放疗敏感性呈显著负相关,这表明PKM2的表达可作为预测非小细胞肺癌患者放疗敏感性的重要分子标志,从而实现非小细胞肺癌患者的个体化治疗.
