祛风宣痹方对支气管哮喘大鼠肺组织内核因子-κB表达的影响

         

摘要

目的 探讨祛风宣痹方治疗支气管哮喘的作用机制.方法 将清洁级雄性SD大鼠50只随机分为5组,即空白组、模型组、泼尼松组、祛风宣痹方低剂量组及祛风宣痹方高剂量组,各10只.采用免疫组织化学染色技术,即二氨基联苯胺(DAB)显色检测大鼠肺组织内核因子-κB p65蛋白(NF-κB p65);计算机图像分析技术对肺组织NF-κB p65表达进行定量检测.结果 模型组大鼠肺组织构成NF-κB阳性细胞网络,较空白组比较NF-κB细胞密度显著增加(P<0.01);泼尼松组、祛风宣痹方高剂量组肺组织NF-κB细胞密度较模型组显著下降(P<0.01),但祛风宣痹方低剂量组无明显下降(P>0.05).结论 祛风宣痹方有效治疗哮喘的作用与抑制NF-κB活化密切相关.%Objective To study the action mechanism of Qufeng-xuanbi formula on the treatment of bronchial asthma.Methods 50 male SD rats (clean class) were randomly divided into five groups,that blank group,model group,prednisone group,Qufeng-xuanbi formula low dose group (low dose group) and Qufeng-xuanbi formula high dose group (high dose group),10 rats in each group.The immunohistochemical staining method,namely,DAB-staining technique,was used to detect nuclear factor-kappa B P65 (NF-κB p65).The expression of NF-κB p65 was detected by computer image analysis system.Results All NF-κB p65 staining in model group could be accounted for a net work,and the density of NF-κB p65 staining was higher than that in control group (P <0.01).The prednisone and high dose group had a significantly lower density of NF-κB p65 staining than that of model group (P < 0.01),but with no obviously decreased as compared with that in low dose group (P > 0.05).Conclusion The therapeutic action of Qufeng-xuanbi formula on bronchial asthma may related to the decrement of activated NF-κB cells.
