结直肠癌组织Toll样受体4和髓系分化蛋白88mRNA检测

摘要

目的 通过检测结直肠癌组织Toll样受体4(TLR4)mRNA和髓系分化蛋白88(MyD88)mRNA,并分析其与分化、分期、转移的相关性,阐明TLR4和MyD88在结直肠癌发生发展中的作用.方法 实时荧光定量PCR(RT-qPCR)检测62例结直肠癌和25例癌旁正常组织TLR4mRNA和MyD88mRNA.结果 结肠癌组织TLR4 mRNA、MyD88mRNA表达量明显高于癌旁组织(P＜0.01).低分化型结肠癌MyD88mRNA高于高、中分化结肠癌(P＜0.01).Dukes'A、B期TLR4mRNA和MyD88mRNA表达量低于C、D期(P＜0.01).结论 结直肠癌TLR4、MyD88mRNA表达增加,并参与肿瘤的发生发展.%Objective To study the relationship between the increased expression of Toll-like receptors (TLR4) and myeloid differentiation factor 88 (MyD88) mRNA in colorectal cancer tissue and the differentiation,stage and metastasis of colorectal cancer. Methods specimens of colorectal cancer and normal tissues adjacent to colorectal cancer were examined for TLR4mRNA and MyD88mRNA with the method of real-time qPCR Results The TLR4mRNA and MyD88mRNA in eolorectal cancer tissue were significantly higher than those in tissues adjacent to colorectal cancer (P＜0.01). In addition, MyD88mRNA was significantly associated with differentiation stares (P＜0.01). Both MyD88mRNA and TLR4mRNA were significantly associated with Duels stage (P＜0.01), but was not associated with lymph node metastasis. Conclusion Increased expression of TLR4mRNA and MyD88mRNA in colorectal cancer tissue may play an important role in the carcinogenesis and progression of colorectal cancer.
