HOXB6在裸鼠大肠癌肝转移模型抑癌机制的实验研究

             

摘要

目的 研究HOXB6在裸鼠大肠癌肝转移模型抑癌机制及其前临床意义.方法 采用基因重组质粒pcDNA6-HOXB6转染的HT29大肠癌细胞株建立裸鼠肝转移模型,随机分为两组,即HOXB6组和对照组.制模30 d后观察两组间转移瘤形成情况及生存情况.结果 HOXB6组平均体质量、肝质量、肝转移评分及血性腹水生成率明显低于对照组,差异均有统计学意义(分别为20.04 g与23.08 g,P<0.01;1.54 g与2.22 g,P<0.01;0.60与3.80,P<0.01;0与80.0%,P<0.05);两组生存曲线分析显示,HOXB6组生存率明显高于对照组,差异有统计学意义(P<0.05).结论 HOXB6可抑制裸鼠大肠癌肝转移的形成,且可作为肿瘤转移的评判指标.%Objective To study the tumor-suppressive mechanisms of HOXB6 gene in nude mice models of liv-er metastasis of colon cancer and its preclinical significance. Methods HT29 colon cancer cells transfected by recom-binant vector pcDNA6-HOXB6 gene were used to establish nude mice models of liver metastasis of colon cancer. The models were divided into two groups: HOXB6 group and control group. The formation situation of metastatic tumor and the survival status of nude mice models in the two groups were studied 30 d after the establishment of models. Results The mean body weight, liver weight, liver metastasis scores and bloody ascites rate of HOXB6 group were sig-nificantly less or lower than those in the control group (20.04 g vs 23.08 g, P<0.01;1.54 g vs 2.22 g, P<0.01;0.60 vs 3.80, P<0.01; 0 vs. 80.0%, P<0.05). Survival curves showed that the survival rate in HOXB6 group was significantly higher than that in the control group (P=0.034). Conclusion HOXB6 gene may inhibit the formation of liver metastasis of colon cancer in nude mices, and it can be regarded as an indicator for evaluating tumor metastasis.
