茶子未成熟胚子叶柄离体培养诱导再生植株

     

摘要

Results from in vitro culture and plant regeneration of cotyledon petiole of immature embryo from Camellia sinensis, aiming at increasing the individual number of hybrid Camellia sinensis rapidly,show that the imature embryo can normally germinate on the two media, MS+BA 3 mg/L+IBA 2 mg/L+ YE 200 mg/L and improve ER+BA 2.5 mg/L+NAA 0.2 mg/L; Callus can be induced and regenerated on medium of improve ER+BA 2. 5 mg/L+NAA 0. 2 mg/L. The propagation effect of callus with bud (propagation time 5.4, suitable culture period 39 d) is significantly better than stem segment with axillary bud, the effective seedling rate is up to 83% with strong and tidy shoots. The optimum medium for radication is 1/2 improved ER+IBA 0.1 mg/L.%为迅速提高茶树杂交后代群体的个体数,进行了茶树未成熟胚培养获得芽苗及子叶柄,再以芽苗和子叶柄为外植体诱导再生植株的试验.结果表明:MS+BA 3 mg/L+IBA 2 mg/L+YE 200 mg/L和改良ER+BA 2.5 mg/L+NAA 0.2 mg/L两种培养基均能较好地诱导未成熟胚正常萌发;改良ER+BA 2.0 mg/L+NAA 0.1 mg/L培养基可较好地诱导子叶柄产生愈伤组织并分化出芽;以培养带芽愈伤组织块方式增殖效果明显比用带腋芽茎段增殖方式好:增殖倍数为5.4,继代周期39 d,芽苗健壮整齐,有效苗率达83%;芽苗生根的最适培养基为1/2改良ER+IBA 0.1 mg/L.
