透明质酸对大鼠颞下颌关节滑膜细胞wnt/β-catenin信号通路的影响

     

摘要

Objective To observe the expression of classic Wnt signaling pathwayβ-catenin in the rat temporomandibular synovial cell and regulation of hyaluronic acid on rat temporomandibular joint(TMJ) osteoarthritis (OA) synovial cell nucleus in β-catenin and MMP-9. Methods Create temporomandibular joint osteoarthritis model in rats with Lekkas method. Primary culture normal and OA synovial cells with collagenase digestion. Cells were divided into normal group, OA group, and hyaluronic acid group. Hyaluronic acid acted on all of cells. MMP-9, nuclearβ-catenin protein expression were assayed. Results OA synovial cells were successfully cultured. Western blot showed that, MMP-9, nuclearβ-catenin expression was significantly higher in OA synovial cells than normal synovial cells. Hyaluronic acid downregulated MMP-9, nuclearβ-catenin expression in OA synovial cells. OA synovial cell supernatant MMP-9 expression was significantly higher than the normal. Hyaluronic acid exert significant downward effect. Conclusion Hyaluronic acid significant downregulated nucleusβ-catenin in OA synovial cells, indicating that hyaluronic acid can regulate Wnt/β-catenin signaling pathway.%目的:观察Wnt经典信号通路中β-catenin在大鼠颞下颌关节炎滑膜细胞胞核中的表达,并观察透明质酸对大鼠颞下颌关节炎(OA)颞下颌关节滑膜细胞胞核中β-catenin和滑膜细胞MMP-9的调控作用。方法采用Lekkas法建立大鼠颞下颌关节骨性关节炎模型,将所培养滑膜细胞分为正常组、OA模型组和透明质酸组,用透明质酸作用于滑膜细胞,检测滑膜细胞中MMP-9、胞核β-catenin的蛋白表达变化及上清液中MMP-9表达变化。结果 Western Blot结果显示OA滑膜细胞中MMP-9、胞核β-catenin表达均明显高于正常滑膜细胞,透明质酸对OA滑膜细胞中MMP-9、胞核β-catenin表达有明显下调作用;OA滑膜细胞上清液中MMP-9表达明显高于正常滑膜细胞上清液,透明质酸对其有明显下调作用。结论透明质酸可以下调颞下颌关节OA滑膜细胞胞核β-catenin的表达,这说明透明质酸对颞下颌关节OA滑膜细胞Wnt/β-catenin信号通路具有一定的调控作用。
