低温冷藏时间对医用级丝光绿蝇幼虫存活率及发育的影响

     

摘要

目的 探讨低温冷藏时间对医用级丝光绿蝇幼虫存活率及发育的影响.方法 采集室内传代培养的丝光绿蝇蝇卵,经消毒后孵化出1日龄、2日龄、3日龄幼虫,再次消毒后于6℃保存,计算冷藏保存第1 ~7 天各日龄幼虫的存活率,存活率>50%时取存活的蛆虫于常温下继续培养2 d,记录蛆虫的存活率及体重.结果 1日龄蛆虫冷藏第1~2天的存活率均>50% ,且高于冷藏第3~7天的蛆虫(均P<0. 05);2日龄蛆虫冷藏第1~3天的存活率均>50% ,且高于冷藏第4~7天的存活率(均P<0. 05);3日龄的蛆虫第1~7天的存活率均>50% ,且冷藏第6~7天存活率均低于冷藏第1~4天的存活率(均P<0. 05). 1日龄蝇蛆冷藏1 d后再培养,2日龄蝇蛆冷藏1~2 d后再培养,3日龄蝇蛆冷藏1~5 d后再培养,其存活率均达80%以上,且生长发育情况良好.结论 1日龄、2日龄、3日龄医用级蛆虫相应的适宜冷藏保存时间分别为1 d、2 d和5 d.%Objective To explore the effects of cryopreservation durations on the survival rate and growth of Lucilia sericata medicinal maggots. Methods The eggs of Lucilia sericata,which were reared by passage in laboratory,were collected,from which hatched 1-,2-,3-day-old larvae after disinfection. Then the larvae were disinfected once more for cryopreservation at a temperature of 6℃. The survival rates of larvae aged different days were calculated during the 1st day to 7th day of cryopreservation. When the survival rate was more than 50% ,the survived larvae,which were kept in room temperature,went on rearing for 2 days. The survival rate and the weight of maggots were recorded. Results The survival rates of 1-day-old maggots on the 1st and 2nd day of cryopreservation were more than 50% , and were higher than those during the 3rd day to 7th day of cryopreservation(all P<0. 05);the survival rates of 2-day-old maggots during the 1st and 3rd day of cryopreservation were all more than 50% ,and were higher than those during the 4th day to 7th day of cryopreservation (all P<0. 05);the survival rates of 3-day-old maggots during the 1st to 7th day of cryopreservation were all more than 50% ,and the survival rates on the 6th and 7th day of cryopreservation were higher than those during the 1st day to 4th day of cryopreservation(all P<0. 05). The 1-day-old larvae were reared after 1 day of cryopreservation,the 2-day-old larvae were reared after 1 to 2 days of cryopreservation,the 3-day-old larvae were reared after 1 to 5 days of cryopreservation,and their survival rates were all more than 80% ,achieving favorable growth and development. Conclusion The corresponding appropriate cryopreservation durations for 1-,2-and 3-day-old Lucilia sericata medicinal maggots are 1,2,and 5 days respectively.
