穿心莲内酯联合顺铂对抑制宫颈癌的协同作用

摘要

目的：探讨穿心莲内酯与顺铂联合用药是否具有协同抑制宫颈癌HeLa细胞存活的作用及胱天蛋白酶（Caspase）系统相关的凋亡机制。方法体外培养HeLa细胞株，经穿心莲内酯和（或）顺铂处理后，MTS法检测细胞的存活率，流式细胞仪技术检测细胞凋亡水平变化，蛋白印迹（ Western blot ）方法检测凋亡相关蛋白Caspase 的亚型（Caspase-3、Caspase-8、Caspase-9）和DNA修复酶（PARP）的表达变化。结果穿心莲内酯联合顺铂能够明显抑制HeLa细胞的增殖，其抑制效果明显强于两药单用；穿心莲内酯联合顺铂可以导致HeLa细胞细胞凋亡；两药联用可激活Caspase系统，活化Caspase-3、Caspase-8、Caspase-9，引起PARP蛋白切割。结论穿心莲内酯和顺铂两药联用具有明显的协同作用，能有效抑制HeLa细胞的存活、诱导细胞凋亡，其作用机制可能与诱导凋亡相关信号转导通路Caspase系统活化相关。%Objective To evaluate the synergistic effect of andrographolide and cosplatin on the apoptosis of cer -vical cancer HeLa cells , and to investigate the role of Caspase signal system in this process .Methods After treated by andrographolide and(or) cisplatin, the survival rate of HeLa cells was detected by MTS , while the cellular apoptosis was determined by flow cytometry , and the expression of Caspase -3, -8, -9 and PARP was examined by Western blot . Results As compared with single drug treatment , andrographolide plus cisplatin treatment could significantly reduce the survival of HeLa cells, induce HeLa cellsapoptosis , activate Caspase-3, -8, -9 and thus induce PARP cutting .Con-clusion Andrographolide and cisplatin possess synergistic effects on inhibiting cell proliferation and inducing cell apopto -sis of HeLa cells , which may be related to the activation of the Caspase system .
