大鼠舌鳞癌诱变过程中基质金属蛋白酶13表达变化的研究

     

摘要

Objective To study the expression of transcription level of matrix metalloproteinases 13 (MMP-13) in a rat model of tongue squamous cell carcinoma. Methods Sprague-Dawley rots were randomly divided into three groups.The experimental groups( 16 rats) were given 4-nitroquinoline-l-oxide(4NQO) every day and the control group( 8 rats) was given normal saline. Tongue samples were harvested from experimental rats when they were in epithelial dysplasia and tongue squamous cell carcinoma. The kinetics of MMP-13 expression was assayed by quantity reverse transcriptase-polymerase chain reaction (qRT-PCR). The paraffin-embeded tissue sections were made to observe the tissue pathological change. Results In normal rat tongue, a faint band for MMP-13 was observed by qRT-PCR. The expression of MMP-13 was increased in the tongue of the rats treatead with 4NQO in epithelial dysplasia, but was not statistically significant ( t = - 1. 759, P =0. 129). And the tongue in squamous cell carcinoma rats showed a marked expression, and was statistically significant compared with epithelial dysplasia ( t = -6. 376, P =0. 001 ) and the control rats (normal) ( t =-6. 579, P =0. 001 ). Conclusion The expression of MMP-13 may play an important role in the occurrence of tongue squamous cell carcinoma, and is more remarkable with the cancerous deep expression.%目的 探讨基质金属蛋白酶13(matrix metalloproteinases 13,MMP-13)在大鼠舌鳞癌诱变过程中转录水平表达变化及其临床意义.方法 50只SD大鼠随机抽取10只作为正常对照组,给予正常饮食.另40只每天喂养0.002% 4-硝基喹啉-1-氧化物,于16周时处死20只,选取处于上皮异常增生的8个标本为上皮异常增生组; 24周时处死另外20只,选取发生舌鳞癌的8个标本为鳞癌组.各组利用定量逆转录聚合酶链反应检测MMP-13在不同病变时期舌组织中转录水平的表达变化.结果 大鼠诱癌16周时重度上皮异常增生发生率为75.0%(15/20),大鼠诱癌24周时舌鳞癌发生率为94.8%(18/19).转录水平的检测显示MMP-13上皮异常增生组的表达较正常组上调,但差异无统计学意义(t=-1.759,P=0.129);舌鳞癌组与正常组(t=-6.579,P=0.001)和上皮异常增生组(t=-6.376,P=0.001)比较MMP-13的表达均有统计学意义的上调.结论 MMP-13随癌变的发生表达显著,可能在舌鳞癌的发生过程中起着重要作用.
