摘要

Objective To determine the effect of epigallocatechin gallate (EGCG) and Ni2 + on the expression of cyclinDX and survivin in tongue squamous carcinoma cells (Cal-27). Methods The viability of cells was determined by the MTT reduction assay. Real-time PCR was used to examine the mRNA expression of cyclinDX and survivin. Western blot method was used to analyze the expression level of protein of cyclinDX and survivin. Results The proliferation of Cal-27 cells was significantly inhibited by high concentration of EGCG (greater than 100 μmol/L). The mRNA expression and protein expression of cyclinDX and survivin were also downregulated (P<0. 05). Ni2+ could enhance the inhibition of Cal-27 cells by EGCG. The expression of cyclinDX and survivin were significantly suppressed by EGCG plus Ni2+ (P<(). 05). Conclusions The inhibition of Cal-27 cells and expression of cyclinDX and survivin by EGCG could be enhanced by the mixture of Ni2+ .%目的 研究表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)及镍离子(Ni2+)共同作用对舌鳞癌细胞系Cat-27 cyclinD1和survivin基因表达的影响.方法 MTT法检测Cal-27的细胞存活率;real-time PCR法检测cyclinD1及survivin mRNA水平表达的影响;Western blot法检测cyclinD1和survivin蛋白水平的表达情况.结果 高浓度EGCG(> 100 μmol/L)可明显抑制Cal-27生长,且对细胞cyclinD1和survivin 基因转录及蛋白表达有明显下调作用(P<0.05);Ni2+与EGCG共同作用增加了对Cal-27的抑制作用,cyclinD1和survivin表达更加显著下降(P<0.05).结论 Ni2+与EGCG共同作用后可增强EGCG对细胞生存率及cyclinD1和survivin基因表达的抑制作用,两者起协同作用.
