HPLC-ELSD法检测蜂蜜饮料中4种可溶性糖含量

     

摘要

建立一种用高效液相色谱-蒸发光检测法测定蜂蜜饮料中葡萄糖、果糖、蔗糖、麦芽糖的检测方法。样品经乙腈-水提取,过滤膜后上机检测。采用ZORBAX糖分析色谱柱(4.6 mm×250 mm,5μm)分离,以乙腈︰水(30︰70,体积比)作为流动相,用蒸发光检测器检测,外标法峰面积定量。结果表明,4种糖组分在0.05 g/100 mL~1.00 g/100 mL浓度范围内线性良好,相关系数r2为0.993~0.997,检出限在0.005 g/100 mL~0.010 g/100 mL,定量限为0.015 g/100 mL~0.030 g/100 mL,加标回收率达到89.4%~97.5%。该方法具有前处理简单、灵敏度高和检测速度快的优点,适用于蜂蜜饮料中葡萄糖、果糖、蔗糖和麦芽糖的定量检测。%A rapid and effective method was established for the determination of glucose, fructose, sucrose, maltose in honey drink by high performance liquid chromatography-ELSD detection method. The samples were extracted by acetonitrile-water,and detected on the machine after filtering membrane. The separation of targeted compound was performed on a ZORBAX sugar analysis chromatography column (4.6 mm×250 mm, 5μm) using acetonitrile:water (30∶70,volume ratio) as mobile phase, with ELSD detector and external standard method peak area quantification. The linear range of four kinds sugar was in the range of 0.05 g/100 mL-1.00 g/100 mL with a correlation coefficient of 0.993-0.997. The detection limit was 0.005 g/100 mL-0.010 g/100 mL and the quantitative limit was0.015 g/100 mL-0.030 g/100 mL. The recovery rate was 89.4%-97.5%. The method has the advantages of simple pretreatment, good purifying effect, high sensitivity and fast detection rate, and is suit-able for the quantitative detection of glucose, fructose, sucrose and maltose in honey drink.
