提高固态发酵培养基灭菌效率的研究

摘要

This research is aimed to solve the "tailing" phenomenon in the sterilization of solid state medium. After analysis the bacterium contamination in the materials of solid state medium, the activation method of spore germination was applied. The results indicated that the sterilization efficiency of solid state medium was able to be elevated remarkably after the pretreatment of heating activation (1. 5% 100 mmol/L L-alanine, 65 ℃ 20 min) , and the quality of solid state medium for culture growth was not affected.%为解决固态发酵培养基的灭菌拖尾问题,在分析固态培养基原料的杂菌污染情况的基础上,采用促进芽孢萌发的方法对培养基进行预处理.结果表明,在培养基中添加1.5％的100 mmol/L L-丙氨酸之后,经过65℃热激活20 min的促芽孢萌发处理可以明显提高培养基的灭菌效率,消除拖尾现象,并且不影响菌种的培养质量.
