滤纸片低温冷冻法保存微生物菌株的临床应用

摘要

Objective To explore the effects of the preservation of microbial strains by filter paper film freezing method. Methods The purified strains were dipped by filter paper, packed with centrifugal tube and freezing preserved at -20℃. Results Among the strains whose storeage time was 6, 12, 18, 24 months respectively, all the survival rate of Pseudomonas aeruginosa, Acinetobacter bauman, Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, Enterococcus faecium, Candida albicans and three standard strains all reached 100%. The morphological characters of the colonies had no obvious changes and bacteria grow well. Conclusion Preservation of strains by low temperature freezing with filter is simple and economic. It is suitable to save non fastidious bacteria in large quantities for clinical laboratory.%目的：观察滤纸片低温冷冻法保存临床微生物菌株的保存效果。方法用滤纸片沾取需保存的纯化菌株，分装离心管中置-20℃低温冷冻保存。结果保存期分别为6、12、18、24个月的菌株中，铜绿假单胞菌、鲍曼不动杆菌、大肠埃希菌、肺炎克雷伯菌、金黄色葡萄球菌、屎肠球菌、白念珠菌以及3株标准菌株存活率均达到100％，菌落形态特征无明显改变且生长良好。结论滤纸片低温冷冻法保存微生物菌株简单、经济，适合临床实验室大批量保存非苛养菌。
