肾复康汤剂对IgA肾病患者外周血T淋巴细胞体外作用研究

摘要

Objective To investigate the influence of shenfukang prescription on activation of T lymphocyte in peripheral blood of IgA nephropathy patients. Methods The CD25, CD69 and HLA-DR expression of CD3+CD4+T and CD3+CD8+T cells of 75 cases of IgA nephropathy patients and 38 healthy control groups were detected. The CD3+CD4+T and CD3+CD8+T cell of IgA nephropathy patients were disposed with PBS, 10 μg/ml, 20 μg/ml, 40 μg/ml, 80 μg/ml of shenfukang, and then to observe the expression changes of CD25, CD69 and HLA-DR. After stimulation of PHA, the CD25, CD69 and HLA-DR expression changes of CD3+CD4+T and CD3+CD8+T cells were recorded. Results The expression of HLA-DR on CD3+CD4+T and CD3+CD8+T cells in IgA nephropathy patients were significantly higher than the normal controls (t=5.03, 5.87, P<0.05). The differences of CD25,CD69 on CD3+CD4+T and CD3+CD8+T cells between two groups had not statistical significance (t=1.43, 1.64, 1.91, 1.69, P>0.05). The CD25, CD69 and HLA-DR expression of CD3+CD4+T and CD3+CD8+T cells of different concentration of shenfukang groups and PBS control group were not significantly different (F=0.19,0.49,0.20,0.59,0.78,0.51,P>0.05) . Conclusions The effect of shenfukang prescription on renal protecting in IgA nephropathy patients may be related with reducing the hidden activation ability of T lymphocyte in peripheral blood.%目的：观察肾复康汤剂体外对IgA肾病患者外周血T淋巴细胞活化功能的影响。方法检测75例IgA肾病患者和38例对照组CD3+CD4+T和CD3+CD8+T细胞上活化指标CD25、CD69、HLA-DR的表达，并将PBS、10μg/ml、20μg/ml、40μg/ml、80μg/ml的肾复康汤剂作用于 IgA肾病患者的CD3+CD4+T和CD3+CD8+T细胞，观察CD25、CD69、HLA-DR的表达变化。结果 IgA肾病患者组中CD3+CD4+T和CD3+CD8+T细胞上HLA-DR的表达率均高于对照组，差异均有统计学意义（t分别=5.03、5.87，P均＜0.05）。两组中CD3+CD4+T和CD3+CD8+T细胞上CD25、CD69的表达比较，差异均没有统计学意义（t分别=1.43、1.64、1.91、1.69，P均＞0.05）。不同浓度肾复康汤剂和PBS孵育对照组间的CD3+CD4+T细胞和CD3+CD8+T细胞上的CD25、CD69、HLA-DR表达率比较，差异均无统计学意义（F分别=0.19、0.49、0.20、0.59、0.78、0.51，P均＞0.05）。结论肾复康汤剂对IgA肾病患者肾脏的保护作用可能和其能降低外周血T淋巴细胞的活化潜能有关。
