高渗盐水联合牛磺酸预处理对肝脏缺血再灌注损伤的保护作用

             

摘要

目的 探讨高渗盐水联合牛磺酸预处理对大鼠肝脏缺血再灌注损伤的保护作用及其作用机制.方法 将30只SD大鼠随机等分为5组:假手术组(A组)、缺血再灌注组(B组)、高渗盐水预处理组(C组)、牛磺酸预处理组(D组)和高渗盐水联合牛磺酸预处理组(E组).参照Pringle法建立大鼠肝脏缺血再灌注损伤模型.再灌注6 h后检测血清谷丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST) 、乳酸脱氢酶(LDH) 、超氧化物歧化酶(SOD)和丙二醛(MDA)水平,并在光镜下观察肝组织病理学的改变和肝脏的原位细胞凋亡的变化.结果 缺血再灌注6 h后血清ALT、AST、LDH 和MDA水平比较,B、C、D和E组明显高于A组(P<0.01),C、D和E组明显低于B组(P<0.01),C组和D组明显高于E组(P<0.05);血清SOD水平比较,A、C、D和E组明显高于B组(P<0.01),E组明显高于C组和D组(P<0.01);肝组织病理学显示B组损害最重,C、D组次之,E组最轻.TUNEL法检测细胞凋亡显示大鼠肝细胞凋亡率B、C、D和E组明显高于A组(P<0.01),C、D和E组明显低于B组(P<0.01).结论 高渗盐水和牛磺酸联合应用可以改善大鼠肝脏缺血再灌注损伤.%Objective To study the protective effect and its mechanism of hypertonic saline and taurine pretreatment on hepatic ischemia reperfusion injury (HIRI) in rats. Methods Thirty healthy adult male Sprague-Dawley(SD) rats were randomly divided into five groups (n = 6),sham operation group(A),IR group (B) , hypertonic saline pretreatment group(C) , taurine pretreatment group(D) ,hypertonic saline and taurine pretreatment group(E). HIRI model was replicated according to Pringle method. The level of alanine aminotransferase(ALT) ,aspartate aminotransferase(AST) , lactate dehydrogenase (LDH) , malondialdehyde(MDA) and superoxide dismutase(SOD) in serum were measured;The histopathological changes of liver and apoptosis of hepatocytes were observed by lightmicroscopies and TUNEL method,respectively. Results Serum level of ALT, AST,LDH and MDA in group B,C,D and E were significantly higher than those in group A(P<0. 01) ;in group CD and E were significantly lower than in group B(P< 0. 01) ,but the level of SOD were significantly higher than in group B(P<0. 01). Conclusion Hypertonic saline combined with taurine can protect liver from ischemia reperfusion.
