甘露糖结合凝集素基因多态性与肺结核的相关性研究

     

摘要

目的 探讨甘露糖结合凝集素(MBL)基因多态性与肺结核的相关性.方法 收集新乡医学院第一附属医院肺结核患者158例作为研究组,其生物学父母作为对照组.运用聚合酶链反应扩增及单核苷酸多态性的分子生物学技术,对MBL基因的rs69287125、rs137824326多态性分型,进行肺结核与甘露糖结合凝集素基因多态性的关联分析和单体型相对风险率分析.结果 rs69287125等位基因与肺结核相关联(P=0.011),其中等位基因G是保护因素(Z=-2.31),A为危险因素(Z=2.31);rs137824326等位基因与肺结核相关联(P=0.007),其中等位基因G是保护因素(Z=-2.54),A为危险因素(Z=2.54).rs69287125-rs137824326单体型的A/A、G/G与肺结核有关联(P=0.021,0.015;Z=2.16,―1.85).结论 MBL基因与肺结核相关联.%Objective To detect the association between pulmonary tuberculosis and gene poly morphism of mannose binding lectin (MBL). Methods The study group included 158 patients with pulmonary tuberculosis and the control group were there par-ents. The polymorphism of rs69287125、rsl37824326 of mannose binding lectin gene was detected with PCR methods and SNP typ-ing in all nucleus families to analyze the correlation and haplotype relative risk. Results The rs69287125 allele was connected with pulmonary tuberculosis(P = 0.011). Allele G was protecting factor(Z= -2. 31) and allele A was the hazard factor(Z = 2. 31). The rs137824326 allele was connected with pulmonary tuberculosis(P = 0. 007). Allele G was protecting factor(Z= - 2. 54) and allele A was the hazard factor(Z = 2. 54). The haplotypes of A/A and G/G in the rs69287125-rsl37824326 were associated with pulmonary tuberculosis (P = 0. 021,0. 015;Z = 2. 16,- 1. 85). Conclusion It shows an association between pulmonary tuberculosis and the polymorphism at nucleotide of mannose binding lectin gene in Chinese.
