Objective To investigate the expression, clinical significance and correlation with T lymphocytes infiltration of cal-reticulin(CRT) and CD47 in laryngeal squamous cell carcinoma (LSCC ). Methods The expression of CRT and CD47 mRN A in 42 cases of LSCC and 18 cases of peri-cancer tissues was detected by reverse transcription-polymerase chain reaction (RT-PCR),the immunohisto-chemistry was performed to detect the protein level of CRT and CD47 and T lymphocytes infiltration in 68 cases of LSCC and 20 cases of peri-cancer tissues. Results The expression of CRT and CD47 was significantly higher in LSCC than in peri-cancer tissue both in mRN A and protein levels (P<0.05). The expressin of CRT and CD47 protein increased with the increase of T stage, clinical stage and lymph node metastasis of patients with LSCC (P<0. 05 ), CD47 protein was positively correlated with the expression of CRT (r=0. 89, P<0. 05 ), and the expression of CRT protein also related with the high infiltration of CD45RO+ T lymphocytes(P<0. 05 ). Conclusion CRT and CD47 can play an important role in the tumorigenesis and development of LSCC, the exprssion of CRT is associated with infiltration of T lymphocytes, which may be a new anti-tumor immune mechanisms of LSCC.%目的 探讨喉鳞状上皮细胞癌(LSCC)组织中钙网蛋白(CRT)和CD47的表达水平、临床意义及其与T淋巴细胞浸润的相关性.方法 应用逆转录酶-聚合酶链反应(RT-PCR)检测42例LSCC组织及18例癌旁正常鳞状上皮组织中CRT和CD47 mRNA的表达情况,用免疫组织化学S-P法对68例原发性LSCC及20例癌旁正常组织中CRT和CD47蛋白的表达、T淋巴细胞浸润情况进行了检测.结果 在LSCC组织中,CRT和CD47 mRNA和蛋白的表达水平均高于癌旁正常鳞状上皮组织(P<0.05).CRT及CD47蛋白随着LSCC的肿瘤分期、淋巴结转移及临床分期的增加其表达水平增加(P<0.05),CD47与CRT蛋白的表达有一种较强的正相关性(r=0.89,P<0.05),CRT蛋白的表达还与CD45RO+T淋巴细胞的高浸润相关(P<0.05).结论 CRT和CD47在LSCC的发生、发展过程中起重要作用,CRT的表达和T淋巴细胞的浸润相关,可能是喉鳞癌的一种新的抗肿瘤免疫机制.
