同种生精小管匀浆与佐剂注射诱导大鼠免疫性睾丸损害

         

摘要

目的 探讨免疫性睾丸损害导致无精子症的发病机制.方法 给雄性大鼠足垫、背部及睾丸多次分别注射同种异体生精小管匀浆与弗氏佐剂混悬液及结核菌素,诱导免疫性睾丸损害.对实验组及对照组生精小管进行组织学、免疫荧光检查并对管腔液中的化学成分进行检测.结果 实验组睾丸质量及生精小管管腔液中大多数化学成分与对照组差异有统计学意义.实验组生精小管无精子,组织学检查基底膜出现增厚及透明样变性,免疫荧光检查基底膜可见免疫球蛋白及补体沉积.结论 由注射同种异体生精小管匀浆与弗氏佐剂混悬液及结核菌素诱导的免疫性睾丸损害可导致生精小管管腔液成分出现显著变化,生精小管病理改变及生精障碍型无精子症.%Objective To investigate the pathogenesis of azoospermia induced by immunological testicular in-jury. Methods The homogenate of allogeneic seminiferous tubule, Freund′s adjuvant suspension and tuberculin were injected into foot pads, backs and testicles of male rats for multiple times to induce immunological testicular injury. The seminiferous tubules in the study group and the control group were determined by histology and immunofluores-cence examination, and the chemical composition in the luminal fluid was tested. Results There were significant dif-ferences in the testicular weight and the main chemical composition in the luminal fluid of seminiferous tubules be-tween the study group and the control group. There was no sperm in the seminiferous tubules of the study group. The histological examination showed thickening and hyaline degeneration of the basilar membrane. The immunofluores-cence examination showed that there was sediment of immunoglobulin and complement at the basement membrane. Conclusion Immunological testicular injury induced by injection of allogeneic seminiferous tubules, Freund′s adju-vant suspensions and tuberculin may cause significant changes in luminal fluid composition of seminiferous tubules.
