槲皮素抑制诱导型一氧化氮合酶减轻脓毒症大鼠的心肌损伤

         

摘要

Aim To investigate the protective effects ofquercetin ( QCT ) treatment on sepsis - induced myocar-dial depression and explore its mechanism. Methods Male Sprague-Dawley rats were randomized to 1 of 3 treatments: sham operation group, CLP group and CLP pretreatment with QCT (10 rats in each group ). Rat sepsis was created by cecal ligation puncture ( CLP ) operation. Left ventricular ( LV ) performance was measured at 12 hours following CLP operation by cardiac hemodynamic measurements via a catheter placed through right common carotid artery to the left ventricle. The expression of inducible nitric oxide synthase ( iNOS ) and the level of nitric oxide ( NO ), and the activity of iNOS of the myocardium was detected. Results Cardiac function of CLP group rats was de-pressed markedly by measuring the LVSP, LVEDP, and ± dp/di^ at 12 hours following CLP operation. iNOS protein of the myocardium was over-expressed, and the level of nitric oxide ( NO ), the activity of iNOS of the myocardium were also increased. But pretreatment with QCT could weaken all these effects. Conclusion The protective effect of QCT against sepsis-induced myocar-dial depression is associated with inhibiting iNOS over-expression and activity, and decreasing the availability of myocardial NO.%目的 初步探讨槲皮素对脓毒症心肌损伤的影响及其机制.方法 将30只SPF级♂ SD大鼠分为槲皮素预处理组、脓毒症组和假手术组,每组10只.采用盲肠结扎穿刺法制备脓毒症大鼠模型.盲肠结扎穿刺术后12 h经右颈总动脉左心室内插管监测各组大鼠MAP、LVSP、LVEDP、±dp/dtmax等血流动力学指标,免疫印迹法测定心肌组织iNOS蛋白表达,酶法测定心肌组织匀浆中iNOS活力及NO含量.结果 脓毒症组大鼠LVSP降低、LVEDP升高、+dp/dtmax与-dp/dtmax均有所下降,而槲皮素预处理组以上各项指标均有不同程度改善;脓毒症组大鼠左心室心肌组织iNOS的表达上调,iNOS的活力升高,NO的含量增加,而槲皮素预处理组以上指标的增加幅度较小.结论 槲皮素通过降低心肌组织iNOS的表达及活力,抑制脓毒症时心肌组织中过量的NO产生,减轻脓毒症心肌损伤.
