鱼腥草中生物碱类成分及其抗血小板聚集活性

     

摘要

AIM:To study the chemical constituents from Houttuynia cordata and to test their antiplatelet aggregation activities.METHODS:The chemical constituents were isolated and purified by silica gel column chromatography and their structures were elucidated on the basis of spectral analysis.The antiplatelet aggregation activities were evaluated by Born's method,using rat PRP induced by ADP and thrombin.RESULTS:Nine alkaloids were isolated and their structures were identified as norcepharadione B (1),4,5-dioxodehydroasimilobine (2),cepharadione B (3),aristoiolactam B Ⅱ (4),aristololactam A Ⅱ (5),sauristolactam (6),piperolactam A (7),splendidine (8),and aristololactam F Ⅱ (9).Antiplatelet aggregation test indicated significant inhibitory activities of compounds 1,2,4-7,9 induced by ADP and 1,2,4,7 induced by thrombin.CONCLUSION:Compounds 2,6,7,9 were isolated from the genus Houttuynia for the first time.Compounds 1,2,4,7 showed significant inhibitory effects on both ADP-induced and thrombin-induced platelet aggregation.%目的:对鱼腥草中生物碱及其抗血小板聚集活性进行研究.方法:采用硅胶柱色谱,重结晶等方法进行分离纯化,并利用化合物的理化性质和波谱数据鉴定其结构;采用Born比浊法测定所得生物碱对ADP和凝血酶诱导的大鼠血小板聚集的影响.结果:从鱼腥草中分离得到9个生物碱,经鉴定分别为:缺碳金线吊乌龟二酮B(1)、4,5-Dioxodehydroasimilobine (2)、cepharadione B (3)、马兜铃内酰胺B Ⅱ(4)、马兜铃内酰胺A Ⅱ(5)、三白草内酰胺(6)、胡椒内酰胺A(7)、Splendidine (8)、马兜铃内酰胺F Ⅱ (9).抗血小板聚集实验表明,化合物1,2,4~7,9对ADP诱导的血小板聚集有显著的抑制作用,其中,1,2,4,7对凝血酶诱导的血小板聚集也具有显著的抑制作用.结论:化合物2,6,7,9首次从该属分离得到,化合物1,2,4,7对ADP和凝血酶诱导的血小板聚集均有显著的抑制作用.
