红霉素和克林霉素对穿通支原体诱导膀胱移行细胞癌干预作用的实验研究

     

摘要

目的 试图探讨抗生素对穿通支原体(Mycoplasma penetrans,Mpe)诱导免疫缺陷小鼠膀胱移行上皮细胞癌的干预作用,为临床上免疫缺陷患者协同穿通支原体感染诱导膀胱肿瘤的干预提供实验依据.方法 70只ICR小鼠隔日腹腔注射环磷酰胺制备免疫抑制模型,分红霉素、克林霉素干预试验组(n=20)在上行注射0.1mLMpe(6×108-9 /mL)的同时肌肉注射红霉素(30mg/kg·d)或克林霉素(25mg/kg·d),连续保护4d.Mpe非干预对照组(n=20)上行感染相同菌量Mpe,NS对照组(n=10)上行注射相同剂量的NS,实验组在第4,8,18w分批宰杀,取小鼠膀胱组织进行微生物学和超微结构检查,计算各组血清培养菌落数,比较显著性差异,并观察小鼠膀胱超微结构的形态学改变.结果 红霉素和克林霉素干预组小鼠菌落检出率较非干预对照组均有显著降低(P <0.01);电镜结果显示红霉素和克林霉素干预试验组小鼠的膀胱组织超微结构显著改善.结论 红霉素和克林霉素对Mpe诱导膀胱移行上皮细胞癌的发生有干预作用.%The aim of the present study was to investigate the antimicrobial intervention of erythromycin and clindamycin against urinary bladder transitional epithelium cell cancer induced by Mycoplasma penetrans(Mpe) in immune-deficient mice,so as to provide the experimental evidence in intervention of the bladder cancer in immune-deficient patients, who has infection caused by Mycoplasma penetrans.All 70 ICR was render to be immunosuppressive by intra-peritoneal injections of cyclophos phamide(50mg/kg d) every other day.The erythromycin-intervention group and clindamycin-intervention group (n=20) were firstly received 0.1ml Mpe(6 × 108-9/mL) by ascending infection through urethra, then received erythromycin (30mg/kg d)and clindamycin(25mg/kg d) respectively by intramuscular injection for continuous 4 days.Non-intervention group (n= 20) received equal Mpe, and normal saline contrast group(n= 10) received equal stroke-physiological saline solution respectively.The treated mice were killed 4, 8 and 18 weeks later and the bladder tissues were taken for microbiological cells and ultrastructure examination.The number of Mpe cultured in serum in each group was evaluated and compared.The morphological changes of the bladder migratory cells were observed.The detection rate of colony in the intervention groups and non-intervention groups had significant difference (P<0.01).Ultrastructural observation showed the tissue structures of the bladder in intervened mice were improved than those without intervention.It's indicated that the erythromycin and clindamycin could intervene immunedeficient mice against the bladder cancer induced by Mycoplasma penetrans.
