孕激素对脂多糖诱导的蜕膜基质细胞的影响

             

摘要

目的 探讨孕激素对细菌脂多糖诱导的蜕膜细胞活性的影响.方法 获取正常早期妊娠妇女的蜕膜组织,经细菌脂多糖刺激体外培养孕激素作用后,用四氮唑盐比色法测定蜕膜细胞增殖抑制情况.结果 不同浓度的细菌脂多糖均抑制了蜕膜基质细胞的生长,孕激素明显抑制了细菌脂多糖对蜕膜基质细胞生长的影响,以孕激素浓度为0.05μg/mL时蜕膜基质细胞增殖最高(F=5.389,P<0.05).结论 细菌脂多糖抑制早孕蜕膜细胞的生长,孕激素明显抑制了细菌脂多糖对蜕膜基质细胞生长的影响,提示孕激素参与到反对细胞内毒素的保护机制.%Objective To investigate the effects of progesterone on bacterial lipopolysaccharide ( LPS )-induced cell death in cultured human decidual stromal cells. Methods Decidual specimens from elective terminations between 7 and 10 weeks of gestation were obtained and cultured with LPS in vitro. The proliferation inhibition of decidual cells was determined with MTT assay after acted by progesterone.Results We found that LPS treatment induced decidual stromal cell death in a dose-dependent manner and that progesterone bloc:ked the effect of LPS treatment. There was significant proliferation among cells co-incubated with LPS at lOμg/mL and progesterone at 0. 05 μg/mL compared with LPS and progesterone at 0. 01 , 0. 1, 0. 2 and 0. 5μg/mL(P<0. 01 ). Conclusion LPS leads to decidual stromal cell death in vitro but progesterone down-regulates the cell death due to LPS under the same condition, which suggests that progesterone participates a protective mechanism against endotoxin.
