诊断超声辐照大鼠睾丸组织对Fas/FasL蛋白表达的影响

摘要

目的 探讨诊断超声对大鼠睾丸组织生精细胞Fas/FasL蛋白表达的影响.方法 32只SD雄性大鼠随机分为4组:I组(对照组)、II组(辐照10 min组)、III组(辐照20 min组)和IV组(辐照30 min组).应用HP 8500彩色多普勒超声诊断仪对大鼠睾丸组织分别进行不同时间的持续辐照,24 h后处死动物,取睾丸组织应用免疫组织化学方法观察标本中Fas/FasL表达情况.结果 正常对照组睾丸组织同时表达Fas和FasL,其表达率分别为59.96%和67.42%.超声辐照后除10 min组外其他各组Fas和FasL的表达率均明显增高(P<0.001).Fas在20 min组总阳性率为68.79%,在30 min组为74.89%,FasL在20 min组为73.81%,在30 min组为81.72%.其表达率随辐照时间延长而增加(P<0.001),强阳性细胞增加更为明显.结论 诊断超声辐照大鼠睾丸组织20 min以上可引起Fas和FasL高表达,提示可导致细胞凋亡.%Objective To investigate the role of Fas/FasL protein expression in rat testes after diagnostic ultrasound radiation.Methods Thirty-two male Sprague-Danles rats were randomly divided into 4 groups of eight according to different radiating time:group I(bland radiation),group II(10 min),group III(20 min),and group IV(30 min).Rat testes were radiated by HP 8500 cobr Doppler ultrasonography with 7.5 MHz linear phased array transducer.Their testes were removed at 24 h after radiation.Expression of Fas/FasL protein was analyzed with immunohistochemistry.Results The positive rates of Fas/FasL protein were higher in experiment groups except group II than in control group,in which the rate of expression of Fas and FasL were 59.96% and 67.42%respectively.After radiation the positive rate was increased in groups III and IV,Fas expression reaching to 68.79% in group III and 74.89% in group IV,FasL expression reaching to 73.81%in group III and 81.72%in group IV respectively.There was no difference between control group and group I(P>0.05).Conclusions Radiation continuously over 20 minutes by diagnostic ultrasound may result in higher expression of Fas and FasL,and further germ cell apoptosis.
