广西主要杧果资源遗传关系的SSR分析

     

摘要

A total of 14 SSRs were used to amplify 63 alleles from 48 major mango accessions available in Guangxi to analyze their genetic diversity and phylogeny. All the primers produced clear and strong expected amplifications. The number of putative alleles was 3 to 6 per locus with an average number of alleles (A) being 4.5 and an effective number of alleles (Ne) being 2.66. The polymorphism information content (PIC) per locus ranged between 0.192 9 and 0.686 3, averaging 0.533 7. The observed (Ho) and expected(He) heterozygosities averaged 0.424 1 and 0.594 3, respectively. Genetic similarity coefficients of all the accessions ranged from 0.455 9 to 0.941 2, averaging 0.705 3, with the minimum between mango 'Guire 06 -3' and ' Guire 06 -5', and the maximum between ' Keitt' and ' Red Keitt' and between ' Yuwenmang' and ' Mitaomang'. UPGMA cluster analysis grouped the mango accessions to 5 groups according to their origin and their classification reflecting the pedigree of the cultivars. The clustering results showed that interspecific genetic relation of all the mango accessions was not related with the nucellar embryony (either monoembryonic or polyembryonic) and that most of the mango accessions in Guangxi were progenies of crosses and seedlings of cultivars Irwin, Okrong, Carabo and Neelum.%应用SSR技术,对48个杧果品种(系)的遗传关系进行了检测.14个SSR基因座在48个品种(系)中分别扩增出3~6个等位基因,共获得63个等位基因,等位基因数(A)平均为4.50;有效等位基因数(Ne)平均为2.66,各基因座的多态信息含量(PIC)值在0.1929~0.686 3之间,平均0.533 7,平均杂合度观测值(Ho)和期望值(He)分别为0.424 1和0.594 3.品种间的遗传相似系数在0.455 9~0.941 2之间,平均为0.705 3.以桂热杧06-3号与桂热杧06-5号两者之间的相似系数最小,而Keitt与红凯特杧及玉文杧与蜜桃杧之间相似系数最大.UPGMA聚类分析将48个品种分为5大类群.聚类结果表明,在分子水平上,杧果品种间的亲缘关系与胚性无直接关系;且广西本地杧果品种(系)多是象牙杧、青皮杧、吕宋杧或秋杧等的杂交子代或实生后代.
