新型吲哚七甲川菁染料HI-6的合成及光声成像评价

         

摘要

Quaternary ammonium salt(B) was obtained by the reaction of 2,3,3-trimethylindolenine with therahydrofuran methyl bromide.Then B was reacted with 2-chloro-3-hydroxy-methylene cyclo-hexene aldehyde to synthesize the novel heptamethine indocyanine dye( HI-6) with the side chain-furan nucleus.The structure was characterized by 1H NMR,13C NMR and HR-MS( ESI).Compared with ICG, the evaluation of the photoacoustic imaging in vitro and in vivo of BALB/c nude bearing tumour was carried out.The results showed that the maximum absorption wavelength and the maximum emis-sion wavelength of HI-6 were 776 nm and 819 nm, respectively.The in vitro and in vivo photoacoustic signal amplitude of HI-6 was far better than ICG with a certain accumulation in the tumour.%以2,3,3-三甲基-3H吲哚与四氢呋喃甲基溴为原料,制得季铵盐(B); B与2-氯-3-羟次甲基环己烯醛反应合成了以呋喃环为侧链的新型吲哚七甲川菁染料(HI-6),其结构经1H NMR, 13C NMR和HR-MS(ESI)表征.以吲哚箐绿(ICG)为对照,进行了荷瘤小鼠体内/体外的光声成像评价.结果表明:HI-6的最大吸收波长为776 nm,发射波长为819 nm. HI-6在体外和体内的光声强度均远高于ICG,且在肿瘤部位有一定聚集性.
