不同根管冲洗液残余药量抑制粪肠球菌生物膜形成的研究

     

摘要

目的:研究不同根管冲洗液残余药量抗菌活性,为选择抑制粪肠球菌细菌生物膜形成的根管冲洗药物提供实参考依据.方法:制备牙本质片样本60个随机分成6组,分别浸泡于0.9%生理盐水(阴性对照组)、2.5%、5.25%次氯酸钠溶液、17%乙二胺四乙酸(EDTA)、10%柠檬酸溶液和2%氯己定溶液中,随后将样本置于400μl粪肠球菌细菌悬液厌氧培养24h后激光共聚焦显微镜观察细菌生物膜形成情况,Bioimage-L软件统计活菌体积并与阴性对照组相比计算每组冲洗液抑制细菌生物膜形成比率.结果:2%氯己定溶液组活菌数最少能抑制99.9%细菌生物膜形成,次氯酸钠溶液组活菌数多抑制细菌生物膜形成能力最差分别抑制13.3%、18.8%细菌生物膜形成,17%乙二胺四乙酸溶液与10%柠檬酸溶液抑制细菌生物膜形成能力次于2%氯已定溶液组分别能抑制43.8%与45.3%细菌生物膜形成.结论:五种根管冲洗液中2%氯己定抑制粪肠球菌细菌生物膜形成能力最强.%Objective: To evaluate the antimicrobial activity of irrigating solutions residual in dentin against Enterococcus faecalis (E.faecalis) biofilms formation. This study provided experimental evidence for seeking a root canal disinfectant with long antimicrobial activity. Methods: 60 dentin slices were randomly divided into 6 groups with 10 in each group, and treated with 0.9% physiological saline (negative control group), 2.5% and 5.25% sodium hypochlorite solution (NaOCL), 17% ethylene diaminetetra acetate (EDTA), 10% citric acid and 2% chlorhexidine (CHX), respectively. After that, each group was incubated with 400|xl E.faecalis suspension for 24h. All specimens were observed under laser scanning confocal microscopy (LSCM). Biofilm scanned images were analyzed and the viable bacteria were calculated by Bioimage-L. The inhibition percentage of biofilm formation was calculated, compared with negative control group. Results: NaOCL groups showed the lowest residual activity (13.3%、18.8%); similar residual activity were found in 17% EDTA and 10% citric acid group with inhibition of E faecalis biofilm formation 43.8% and 45.3% respectively; 2% CHX group showed greatest residual activity (99.9%). Conclusions: 2% CHX was shown to be the most effective in inhibition of E faecalis Biofilm Formation among five different irrigating agents.
