超滤离心法测定连翘酯苷脂质体包封率

摘要

Objective: To establish a method for determining entrapment efficiency of forsythoside lipo-somes. Methods: Forsythoside liposomes were prepared by a reverse evaporation method, and then the entrapment efficiency was determined by ultrafiltration. Results: The free drug was separated from the forsythoside liposomes efficiently. Average recovery rate of free forsythoside was 98. 75% ~ 101. 71% , and the recovery rate was 95. 67% ~ 98.76% in the process of ultrafiltration. The entrapment efficiency of three lot samples was 72. 77% ~ 74. 07% with RSD of 0.56% -1.77% by ultrafiltration. Conclusion: The ultrafiltration method can simply and immediately measure entrapment efficiency of forsythoside liposomes after preparation.%目的:建立连翘酯苷脂质体包封率的测定方法.方法:通过逆相蒸发法制备连翘酯苷脂质体,运用超滤离心法测定其包封率.结果:超滤离心法能快速有效的对脂质体与游离药物进行分离,游离药物平均回收率在98.75％～101.71％之间,超滤加样回收率在95.67％～98.76％之间;此法测定3批样品,所得包封率为72.77％～74.07％,RSD为0.56％～1.77％.结论:超滤离心法操作简单、快速,可用于连翘酯苷脂质体包封率的测定.
