Objective To investigate the expression and significance of S100 alpha 6 protein (S100A6) in the brain of rat model of traumatic brain injury (TBI). Methods Fifty-four male SD rats were divided randomly into normal control group (n -6) and TBI group (n =48). The TBI model was created by using lateral head rotation device and subdivided into 1 h,3h, 6h, 12 h, 24h , 72h, 7d and 14 d group (6 rats in each group). The expression and distribution of S100A6 in the rat brains were investigated by immunohistochernisrry. The expression of the brown stained particles was considered as "positive" and lack of the stained particles as "negative". In order to estimate the contribution of the non-specific interaction binding, a negative antibody control ivas used in each serial sections. Results S100A6 was distributed on nucleus envelope, cytoplasm and cell membrane both in the hippocampus and the cortex. The expression of 5100A6 was decreased to the lowest level in the rat brain at 1 h post TBI (P<0.01), then it became stronger gradually and recovered to normal level at 7 d. There was no statistical difference in the expression of S100A6 at 7 d and 14 d compared with the normal control group (P>0.05). Conclusion The lowest expression of S100A6 after TBI indicates that S100A6 may play an important role in the development of brain injury under the different mechanisms.%目的 探讨大鼠创伤性脑损伤(traumatic brain injury,TBI)后脑组织中钙结合蛋白S100alpha 6(S100A6)的表达规律及意义.方法 雄性SD大鼠54只,按随机数字表法分为正常对照组(6只)和TBI组(48只);采用头颅侧向旋转致伤方法制作TBI模型,并按伤后大鼠处死时间分为1,3,6,12,24,72 h、7d,14 d共8个亚组,每组6只.采用免疫组化方法法检测S100A6蛋白在大鼠脑组织中表达规律及分布情况.S100A6阳性结果判断标准以细胞浆内出现棕黄染色颗粒为阳性,反之为阴性,为消除背景染色的影响,同一组织连续切片设阴性对照组.结果 大鼠脑组织中S100A6蛋白在皮层与海马组织细胞的核膜、胞质、细胞膜中均有表达,脑损伤后一小时时间点皮层与海马细胞表达均最低(P＜0.01),随后表达量逐渐递增,至损伤后7d其表达量恢复正常值,7d、14 d与正常对照组比较,差异无统计学意义(P＞0.05).结论 在TB1后,S100A6蛋白出现一个表达低峰,提示在不同机制影响下可能对脑损伤的发生与发展发挥重要作用.
