新疆栽培荒漠肉苁蓉粗多糖肌肉免疫对OVA蛋白免疫效果的影响

摘要

Objective To investigate the immunopotentiating effects of cultivated Cistanche deser-ticola (C.deserticola) crude polysaccharides (CCDCP) as an adjuvant on the model antigen ovalbumin (OVA). Methods Low,medium and high doses of CCDCP in combination with OVA were intramuscularly injected twice into ICR mice at an interval of two weeks,respectively. Aluminum adjuvant was used to set up positive control group. Levels of IgG,IgG1 and IgG2a antibodies were detected by ELISA. Splenocyte prolif-eration was detected by MTT assay. Growth conditions of the immunized mice were observed. Results IgG level was significantly increased in the high dose group 7 days after the first immunization(P<0.05),espe-cially on the 21st and 28th days (P<0.01) as compared with that of the aluminum adjuvant group. High dose of CCDCP in combination with OVA significantly up-regulated the levels of IgG1 and IgG2a in mice as compared with immunization with OVA alone (P<0.05). Moreover, IgG2a level in mice immunized with high dose of CCDCP and OVA was higher than that of the aluminum adjuvant group(P<0.05). Splenocyte proliferation was significantly enhanced in the medium and low dose groups in comparison with that of the OVA group (P<0.05) and the aluminum adjuvant group (P<0.01). No significant difference in mouse body weight was observed in different groups(P>0.05). Conclusion CCDCP as an adjuvant of OVA pro-tein vaccine can enhance Th1 and Th2 immune responses,especially the early antibody production and Th1 immune response. These results will provide some information for further studies of CCDCP as a vaccine ad-juvant.%目的 探讨新疆栽培荒漠肉苁蓉粗多糖(cultivated C. deserticola crude polysaccharides,CCDCP)作为模式抗原卵清白蛋白(ovalbumin,OVA)佐剂的免疫增强作用.方法 将低、中、高3 个剂量的CCDCP 分别配伍OVA 蛋白,肌肉免疫小鼠,免疫2 次,间隔2 周,阳性对照为铝佐剂组,ELISA法检测小鼠血清中的IgG 及其分型IgG1、IgG2a 抗体的表达水平;MTT 法检测脾细胞增殖;并观察免疫后小鼠的生长状况.结果 高剂量CCDCP 组在免疫后7 d 就可以显著提高IgG 的水平(P<0. 05),在免疫后的21 d 和28 d 可极显著提高IgG 的抗体水平(P<0. 01),与铝佐剂组相比差异有统计学意义(P<0. 05);高剂量CCDCP 组可以显著提高IgG1 和IgG2a 的抗体水平(P<0. 05),与铝佐剂组相当;中剂量和低剂量的CCDCP 显著增强脾细胞的增殖水平(P<0. 05),与铝佐剂组相比差异有统计学意义(P<0. 01);CCDCP 对小鼠体重的影响差异无统计学意义(P>0. 05).结论 CCDCP 作为OVA 蛋白的佐剂能够增强Th1 型和Th2 型的免疫应答,尤其可以诱导早期抗体的产生和Th1 型的免疫应答,为CCDCP 作为疫苗佐剂的研究提供参考.
