瑞舒伐他汀改善胰岛素抵抗的内皮细胞功能的研究

         

摘要

Objective To study rosuvastatin improve the endothelial function on endothelial cells of insulin resist-ance,and the mechanisms to confront atherosclerotic endothelial cells. Method Rosuvastatin as study drug,to estab-lish endothelial cells insulin resistance mode with cell culture,using nitroreductase method for the determination of the content of nitric oxide,using radioimmunoassay for endothelin levels,using western blot to observe the expression of nitric oxide synthase and quantitative analysis. Result Rosuvastatin acting on insulin resistance endothelial cells after 48h,there was a significant difference on NO increasing and ET-1 lowering(P<0. 05);compared with the con-trol group,rosuvastatin significantly increased the intracellular eNOS content(P<0. 05),the optimal concentration of rosuvastatin statin was 10-3 mmol/L. Conclusion Rosuvastatin improve endothelial function of insulin resistance en-dothelial cells significantly,the mechanism may be improving eNOS activity,leading to an increased of NO content and improving endothelial function.%目的:研究瑞舒伐他汀对胰岛素抵抗的内皮细胞的影响及对抗内皮细胞粥样硬化的作用机制。方法采用瑞舒伐他汀作为研究药物,通过细胞培养等方法建立内皮细胞胰岛素抵抗模型,采用硝基还原酶法测定一氧化氮( nitric oxide,NO)的含量,采用放射免疫方法测定内皮素含量,采用western blot方法观察内皮型一氧化氮合酶( endothelial nitric oxide synthase,eNOS)的表达及定量分析。结果瑞舒伐他汀作用于胰岛素抵抗的内皮细胞48小时后,对 NO升高及内皮素-1(endothelin-1,ET-1)降低差异均有显著性(P<0.05);与对照组相比可显著提高细胞内 eNOS 的含量(P<0.05),瑞舒伐他汀最佳浓度为10-3mmol/L。结论瑞舒伐他汀对改善胰岛素抵抗内皮细胞的内皮功能作用显著,其机制可能是通过改善eNOS活性,进而增加NO含量而改善内皮功能。
