珍菊降压片与氨氯地平片合用对大鼠心肌内皮源性血管活性物质昼夜节律的探讨#

         

摘要

Objective:To explore Zhenjvjiangya tablet combination with amlodipine tablet on myocardial endothelium-derived vasoactive substances influence the circadian rhythm. Methods: 64 healthy SD rats, 10 weeks old, weighing 250 to 300 g, male and female, in groups of 32, were randomly divided into two groups(each large group was divided into four groups, each group of 8):(1) control group:amlodipine tablets given to 2.5mg/(kg.d) gavage. (2) experimental group:gavage amlodipine tablets, based on the early, to give Zhenjvjiangya tablet to 40mg/(kg.d) gavage.Continuous administration of 4 weeks. Radioimmunoassay and nitrate reductase assay rats myocardial endothelin-1(ET-1) and nitric oxide(NO) levels.Results:Control group and the experimental group after treatment of myocardial tissue NO rhythm values were increased, ET-1 rhythm curve median decrease(P<0.05), the control group changed little. Compared with before treatment, after treatment, both groups myocardial ET-1 and NO rhythm curve peak phase shift occurs(P<0.05), the ET-1 and NO rhythm changes.Conclusion:Zhenjvjiangya tablet and amlodipine tablets compared to amlodipine alone significantly increased NO levels can reduce the ET-1, control blood pressure and protect the myocardium.%目的:探讨珍菊降压片与氨氯地平片合用对大鼠心肌内皮源性血管活性物质昼夜节律的的影响。方法:健康SD大鼠64只,10周龄,体重250~300克,雌雄各半,按每组32只,随机分为两大组(每大组又分4小组,每小组各8只):(1)对照组:给予氨氯地平片以2.5mg/(kg.d)灌胃。(2)实验组:在灌服氨氯地平片基础上,给予珍菊降压片以40mg/(kg.d)灌胃。持续给药4周。应用放射免疫法及硝酸还原酶法检测大鼠内皮素-1(ET-1)和一氧化氮(NO)水平。结果:对照组和实验组大鼠治疗后的心肌组织NO节律中值均增加,ET-1节律曲线的中值减小(P<0.05),对照组变化幅度较小。与用药前相比,用药后两组大鼠心肌ET-1和NO节律曲线峰值相位发生移位(P<0.05),使ET-1和NO节律发生变化。结论:珍菊降压片联合氨氯地平片较单用氨氯地平片可以明显升高NO水平,降低ET-1,控制血压及保护心肌的作用。
