邻苯二甲酸二丁酯建立尿道下裂大鼠模型的实验研究

         

摘要

目的:采用邻苯二甲酸二丁酯(DBP)建立尿道下裂大鼠模型,探讨尿道下裂的发病原因。方法:将受孕雌鼠随机分为四组(各10只),对照组(D组)给予等体积大豆油;观察组(A、B、C三组)分别每天给予DBP 500、800、1200mg/kg。观察孕鼠分娩后体重及死亡率,统计仔鼠数目和雄雄性仔鼠体重。在仔鼠出生后(PND)7d观察其尿道下裂发生率和隐睾出现率。对发生尿道下裂仔鼠称重后解剖,评价发育情况。结果:C组孕鼠死亡6只,产仔0只,与D组相比差异显著(P<0.01)。B组孕鼠死亡0只,与对照组无差异(P>0.05);B组生产仔鼠、仔鼠平均体重与D组相比差异显著(P<0.05),A组与D组相比无明显差异。B组生产仔鼠出现明显尿道下裂畸形。A组、B组雄性仔鼠均出现明显隐睾。尿道下裂仔鼠肝脏、肾脏、前列腺、睾丸及附睾脏器系数明显降低,脑垂体脏器系数明显增加。结论:DBP对孕鼠有明显毒性作用。每天DBP 800mg/kg可诱导雄性仔鼠尿道下裂的发生,导致仔鼠肝、肾、生殖系统发育不良。%[ABSTRACT]Objective:To use dibutyl phthalate(DBP) the establishment of hypospadias rat model, to explore the causes of hypospadias. Methods: The pregnant female rats were randomly divided into four groups (10 rats each), control group (group D) given equal volume of soybean oil;observation group (A, B, C three groups) were given daily DBP for 500, 800, 1200mg/kg. Observation and mortality after delivery of pregnant rats body weight, weight of newborn rats and the statistical number of male male offspring rats. In rats after birth (PND) 7d to observe the incidence of cryptorchidism and hypospadias occurrence rate. On the occurrence of hypospadias rat pups after weighing anatomy, developmental evaluation. Results: C group of pregnant rats and 6 rats died, 0 mice were born, compared with the D group had signiifcant difference (P<0.01). B group of pregnant rats and 0 rats died, and the control group had no difference (P>0.05); B Group production of offspring rats, average weight of newborn rats compared with the D group signiifcantly (P<0.05), A group and D group had no signiifcant difference. B Group production of newborn rats appear obvious hypospadias. A group and B group of male offspring rats were obvious cryptorchidism. Transurethral resection of liver, kidney, prostate, hypospadias rat pups in epididymis and testis organ coefifcient obviously reduces, the pituitary gland organ coefifcient increased signiifcantly. Conclusion:DBP has obvious toxic effects on pregnant rats. Every DBP 800mg/kg can induce male rats after hypospadias, lead to liver and kidney, newborn rats reproductive system dysplasia.
