血样保存温度及时间对他克莫司血清药物浓度监测结果的影响

             

摘要

目的:研究血液样本在不同保存温度及放置下对他克莫司血清药物浓度监测结果的影响.方法:选取80名受检对象(均为肾移植术后患者),每人采集6ml静脉血,将每份样本按照不同保存温度分成2组(分别放在冷藏4℃和室温20℃),采用均相酶标记放大免疫检测技术(EMIT)测定保存0h、24h、48h的他克莫司血清药物浓度,将不同温度组中24h与0h、48h与0h的血药浓度结果差值进行t检验,观察血样在不同保存温度,血药浓度测定结果的变化有无显著性差异.结果:血液样本的保存温度对EMIT法的监测结果有显著影响,并且随放置时间的延长出现明显变化.结论:血清药物浓度监测结果受保存温度和放置时间等因素的影响,应严格进行质量控制%Objective: A study about the different Standing Time and Storage Temperature of blood sample influence on the Monitoring Serum Concentration of tacrolimus.Methods:80 Tacrolimus Venous blood samples were collected from renal of post-transplantation patients.Each sample was either stored under cold-stored(4℃) or room temperature (20℃),and was tested after 0,24 or 48 hours using EMIT.We used t test to compare the difference of Serum Concentration,24h with 0h,48h with 0h,in different temperature groups.Results:There was significant difference between samples stored under room and refrigeration temperature using EMIT.and the difference was expanding with the extension of placed time.Conclusion:Monitoring of serum concentration was affected by standing time and storage temperature,we should be strict in quality control.
