Cripto-1蛋白、E-钙粘蛋白和波形蛋白在乳腺癌中的表达及其意义

         

摘要

目的:研究Cripto-1蛋白、E-钙粘蛋白(E-cadherin)和波形蛋白(Vimentin)在乳腺癌中的表达情况及意义,探讨Cripto-1蛋白与上皮-间质转化(EMT)在乳腺癌的侵袭与转移过程中的关系。方法采用免疫组织化学方法检测86例乳腺癌和40例癌旁组织中Cripto-1蛋白的表达及86例乳腺癌中E-cadherin和Vimentin蛋白表达情况,分析Cripto-1、E-cadherin和 Vimentin的蛋白表达与乳腺癌临床病理特征的关系及 Cripto-1、E-cadherin和 Vimentin蛋白表达的相关性。结果在86例乳腺癌和40例癌旁组织中Cripto-1的阳性率分别为66.3%和7.5%,E-cadherin的低表达率和Vimentin的高表达率在乳腺癌中分别为24.4%和54.6%。Cripto-1的表达与HER2阳性、有淋巴结转移和高组织学分级有关(P<0.05);E-cadherin的低表达与患者年龄、有无淋巴结转移和组织学分级有关(P<0.05);Vim-entin的高表达与肿瘤大小、有无淋巴结转移和组织学分级有关(P<0.05)。Cripto-1的表达与 E-cadherin低表达和Vimentin的高表达之间呈显著正相关(P<0.05)。结论 Cripto-1蛋白在乳腺癌中的阳性率高;Cripto-1可能通过调控 E-cadherin和 Vimentin的表达来促进 EMT,进而在乳腺癌的侵袭和转移过程发挥重要作用。%Objective To investigate the significance of Cripto-1,E-cadherin and Vimentin expressions in breast cancer,and to study the significance of Cripto-1 which may play an important role in Epithelial-Mesenchymal Transition (EMT)and its prognosis in breast cancer.Methods Immunohistochemistry (ElivisioTMplus)method was used to de-tect Cripto-1 expression in 86 cases of breast cancer and 40 cases of non-cancerous tissues surrounding tumors,and also detect E-cadherin,Vimentin in 86 cases of breast cancer,Then the association between Cripto-1,E-cadherin,Vimentin expressions and clinical pathological characteristics as well as the correlation between among Cripto-1 ,E-cadherin,Vim-entin expressions were analyzed.Results The positive expression rates of Cripto-1 in breast cancer and non-cancerous tissues surrounding tumors were 66.3% and 7.5% respectively,and the low E-cadherin expression as well as the high Vimentin expression in breast cancer were 24.4% and 54.6% respectively.The expression of Cripto-1 protein was sig-nificantly associated with HER2、lymph node metastasis and Histological grade (P<0.05);the expression of E-cadher-in protein was significantly associated with age、lymph node metastasis and Histological grade (P<0.05);the expres-sion of Vimentin protein was significantly associated with the size of tumor,lymph node metastasis and Histological grade (P<0.05).The expression of Cripto-1 was closely correlated with low expression of E-cadherin (r=0.234;P<0.05)and the high expression of Vimentin(r=0.289;P<0.05).On the other hand,the low expression of E-cadherin was closely correlated with that high of Vimentin(r=0.246;P<0.05).Conclusion The positive rates of Cripto-1 ex-pression were significantly higher in breast cancer.What is more,Cripto-1 may promote EMT through the regulation of the expression of E-cadherin and Vimentin,thereby clearly showing that it plays an important role in invasion and me-tastasis of breast cancer.
