梓醇对2型糖尿病大鼠胰岛细胞的保护作用

Objective To study the protective effects of catalpol on the islet cel s in rats with type 2 diabetes mel itus(T2DM).Methods Eight week old male wistar rats were randomly divided into the blank group(n=10)and the experimental group(n=90).The rats in experimental group were fed with high fat and high sugar diet for 8 weeks,after intraperitoneal injection of streptozotocin (STZ,15 mg/kg),with fasting blood glucose more than 16.7 mmol/L as T2DM molded standard.Sixty diabetic rats were randomly di-vided into model group,metformin group,catalpol low,middle and high dose group.The rats in blank group and model group were fed with distil ed water 5 mL/(kg·d).The rats in metformin group was given metformin 90 mg/(kg·d),The rats in catalpol low,middle and high dose groups were fed catalpol 2.5 mg/(kg· d),5 mg/(kg· d),10 mg/(kg· d).After 12 weeks,the rats fasting glucose,fast-ing serum insulin were measured,and the morphology of islet cel s were observed by Hematoxylin and Eosin (HE)staining.Results Compared with the model group,the insulin secretion index increased significantly in each drug intervention group. HE staining showed that islet cel structure was relatively intac in metformin group,catalpol low,medium and high dose groups. Conclusion Catalpol can protect the islet cel s.%目的：探讨梓醇对2型糖尿病大鼠胰岛细胞的保护作用及作用机制。方法8周龄雄性 Wistar大鼠,随机分为正常组10只,实验组90只。实验组以高脂高糖饮食饲养8周,之后腹腔注射链脲佐菌素(STZ,15 mg/kg),以空腹血糖≥16.7 mmol/L作为2型糖尿病成模标准,将符合2型糖尿病成模标准的60只大鼠随机分为模型组、二甲双胍组、梓醇低剂量组、梓醇中剂量组、梓醇高剂量组。正常组和模型组灌胃蒸馏水[5 mL/(kg·d)],二甲双胍组灌胃二甲双胍[90 mg/(kg·d)],梓醇低、中、高剂量组分别灌胃梓醇2.5 mg/(kg· d)、5 mg/(kg· d)、10 mg/(kg· d)。各组大鼠干预12周后,测定空腹血糖、空腹血清胰岛素(FINS),并计算胰岛素分泌指数(HOMA β),HE染色检测大鼠胰岛细胞形态。结果与模型组比较,各药物干预组大鼠胰岛素分泌指数均有所增加；与模型组比较,HE染色结果显示二甲双胍组,梓醇低、中、高剂量组胰岛细胞结构相对完整。结论梓醇可保护胰岛细胞,促进胰岛素分泌。
