芪桂益脉灵对急性心肌缺血再灌注损伤大鼠模型环氧化酶2的影响

         

摘要

Objective To explore the effect Qigui Yimailing decoction(QYD)on Cox 2 in rat myocardial tissue with acute myocardial ischemia reperfusion.Methods Fifty SD male rats were randomly divided into 5 groups and blank control group(n=10)after 7 days physiological saline irrigation,don’t do any processingthe sham operation group(n=10)after 7 days physiological saline irrigation-take the thoracotomy to exposeleft coronary artery of the rats,threading but do not make ligation,the rest of 30 rats were divided into pure ischemia reperfusion group,QYD low dose group and QYD high dose group,and acute myocardial infarction (AMI)models were caused by ligation of the left anterior descending branch of the coronary artery,after the success of the building,to the physiological saline,QYD (high and smal dose,1.2 g/kg and 0.6 g/kg)respectively,whose irrigation served 7 days in a row during the morning.Ab-dominal aorta in pursuance of 5 mL,after centrifugation,get the upper serum,with double antibody sandwich method enzyme linked immunoassay to determine the expression of cox 2.Results The cox 2 of the pure ischemia reperfusion group were significantly higher than the blank control group’s and the sham operation group’s(P<0.05).Compared with pure ischemia reperfusion group, cox 2 overflow of QYD group significantly reduced in rats serum,and the effect of high dose group was more significant.Conclusion Cox 2 has a high level expression in acute myocardial ischemia reperfusion,which prompted inflammation is involved in acute myocardial ischemia reperfusion,QYD can decrease the cox 2 protein expression level and reduce the pure ischemia reperfusion group (I/R)myocardial inflammation,to protect myocardium.%目的:探讨芪桂益脉灵(QGYML)对心肌缺血再灌注大鼠心肌组织环氧化酶2(COX 2)的影响。方法取 SD大鼠50只,随机分为5组,空白组(10只)灌服生理盐水后不做任何处理,假手术组(10只)灌服生理盐水后,开胸暴露左冠状动脉,穿线但不作结扎,其余30只大鼠分别分成单纯缺血再灌注组、芪桂益脉灵低剂量组和芪桂益脉灵高剂量组。并且结扎冠状动脉左前降支造成急性心肌梗死(AMI)模型,造模成功后,分别给生理盐水、QGYML大小剂量(即1.2 g/kg和0.6 g/kg),每日清晨空腹灌胃给药一次,连续灌服7 d。腹主动脉采血5 mL,离心后取上层血清,采用双抗体夹心法酶联免疫测定 COX 2的表达。结果单纯缺血再灌注组 COX 2水平明显高于空白组和假手术组(P<0.05),芪桂益脉灵组与单纯缺血再灌注组相比,大鼠血清中 COX 2表达减少,其中高剂量组的效果更加显著。结论 COX 2在心肌急性缺血再灌注中均有高水平表达,提示炎症参与了急性心肌缺血再灌注,芪桂益脉灵可降低 COX 2的蛋白表达水平,减轻缺血再灌注心肌炎症反应,对心肌起保护作用。
