银杏内酯B对阿尔茨海默病大鼠海马c-fos、Caspase-3和Caspase-8的影响

         

摘要

目的 探讨银杏内酯B(GB)对阿尔茨海默病(AD)大鼠c-fos、Caspase-3和Caspase-8的影响.方法 将大鼠随机分为正常组、假手术组、模型组和银杏内酯B组,采用 Morris水迷宫检测学习记忆,免疫组织化学分析海马c-fos阳性神经元光密度值,荧光定量PCR和Western blot检测海马Caspase-3 和Caspase-8的mRNA和蛋白水平.结果 5 mg/kg的银杏内酯B可缩短AD大鼠逃避潜伏期,增加穿越平台次数,提高海马c-fos的表达,下调凋亡相关基因 Caspase-3和Caspase-8 mRNA和蛋白表达.结论 银杏内酯B可改善AD大鼠学习记忆能力,这种改善可能与上调AD大鼠海马组织c-fos表达,下调Caspase-3和Caspase-8 mRNA和蛋白水平有关.%Objective To evaluate the efficiency of ginkgobalide B on the expression of c-fos,caspase-3 and caspase-8 in the hippocampus of Alzheimer's disease rats.Methods Rats were randomly divided into four groups:normal group,sham group,model group and Ginkgobalide B group.The Morris water maze (MWM) test was used to assess behaviors of the rats,Imunohistochemical staining was used to test c-fos in the hippocampal CA1.Real-time PCR and western blot were used to examine caspase-3 and caspase-8 mRNA and protein levels.Results MWM test result showed that escape latency was significantly shower and the times of crossing the platform increased.Imunohistochemical staining showed that c-fos increased in the hippocampal CA1 after ginkgobalide B (5 mg/kg) treatment.Real-time PCR and western blot manifested that compared with the model group,caspase-3 and caspase-8 decreased in the hippocampus after ginkgobalide B (5 mg/kg) treatment.Conclusion Ginkgobalide B can improve learning and memory in Alzheimer disease's rats,and its mechanism might be associated with neuronal apoptosis.
