电针对非酒精性脂肪性肝炎大鼠肝组织核因子-κB活性与PPARγ表达的影响

         

摘要

目的:应用电针刺激SD大鼠肝俞、足三里、丰隆、太冲穴,观察其对高脂饮食所致的非酒精性脂肪性肝炎(NASH)大鼠肝组织病理改变的影响.方法:40只SD大鼠随机分为正常组、模型组、电针组和西药组,每组10只.正常组大鼠予普通饮食喂养,模型组、电针组和西药组大鼠均给予高脂饮食12周;12周后正常组、模型组同前喂养,其他两组大鼠分别进行电针和西药熊去氧胆酸(UDCA)治疗,共4周;16周全部大鼠断头处死,用HE染色观察大鼠肝组织光镜下的病理改变;EMSA法测定大鼠肝组织NF(核因子)-κB活性;逆转录聚合酶链反应观察各组大鼠肝组织PPAR(过氧化物酶体增殖物激活受体)γ mRNA的表达情况.结果:模型组和西药组大鼠的NF-κB活性明显高于正常组(P均<0.01)和电针组(P均<0.05).模型组、电针组和西药组大鼠肝组织PPARγ mRNA的表达均较正常组有不同程度减弱(P<0.01),且模型组大鼠的表达明显低于电针组(P<0.01).结论:肝组织NF-κB的活性增强及PPARγ的表达减弱与NASH的发生密切相关;电针可以起到增强大鼠肝组织PPARγ的表达及降低NF-κB的活性作用.电针可能通过抑制NF-κB的活性、激活PPARγ的作用而达到治疗NASH的目的.%Objective: To observe the effects of Electronic Acupuncture on NF-kappa B Binding Activity and the Expression of PPAR gamma Mrna in the livers of rats with non-alcoholic steatosis hepatitis, Acupoints Ganshu ( BL 18) , Zusanli ( ST 36 ) , Fenglong (ST 40) and Taichong (LR 3 ) were needled with electronic acupuncture. Methods: Forty SD rats were randomly divided into four groups of ten each: normal group, model group, Electronic Acupuncture group and the Western Medicine group. The rats of the normal group were fed with common food; while the rats of the other three groups were fed with fat-rich diet for In 12 weeks in total 12 weeks later, the rats of the normal group and the model group were fed as before, while the other two groups were punctured with electric acupuncture and lavaged with UDCA respectively for another 4 weeks. The rats were killed at the end of the 16th week from the beginning of the experiment Serum and liver specimens were collected. Histological specimens were stained with HE and then studied. NF-Kb and PPARγ were tested by EMSA and RT-PCR respectively. Results: NF-kappa B binding activity was obviously higher in the model group and the western medicine group than those in the normal group (P< 0.01) and the electric acupuncture group (P<0. 05) . The expression of PPARγ Mrna of the model group, electric group and western medicine group was weaker than that of the normal group (P <0.01) and the level of PPAR-y Mrna was lower in the livers of the model group than that of electric acupuncture group (P <0. 01) . Conclusion; The increased activity of NF-kappa B might play a vital role in the inflammatory response of NASH. Electronic acupuncture may inhibit NF-kappa B binding activity and enhance the expression of PPARγ Mrna so that it should be broadly used in the treatment of NASH.
