金银花提取物中绿原酸含量测定

摘要

Objective To extract and purify chlorogemc acid from Flos lonicerae, and establish an HPLC method to determinate the content of chrlorogenic acid. Methods Chrlorogenic acid in Flos lonicerae was extracted crudely with water, the crude extracts was isolated and purified with macroporous resin. The content of chlorogenic acid in the purified products were determined by HPLC. Results The average contents of chrlorogenic acid in Flos lonicerae was 4.28%. The average chrlorogenic acid contents in purified products was 16.39%. The calibration curve was linear within the range of 0.5-2.5 ng/mL and the average recovery was 99.46%. Conclusion The technology for the extraction and purification of chrlorogenic acid in Flos lonicerae was simple. The method for determining chrlorogenic acid is accurate and repeatable.%目的 提取纯化金银花中绿原酸,并进行含量测定.方法 采用大孔吸附树脂对金银花提取物中的绿原酸纯化,用高效液相色谱法测定绿原酸的含量.结果 金银花中绿原酸的平均含量为4.28％,纯化物中绿原酸的平均含量达16.39％,绿原酸在0.5～ 2.5 μg/mL范围内浓度与峰面积呈良好的线性关系,平均回收率为99.46％.结论 运用大孔树脂分离纯化绿原酸简单易行,含量测定方法精确,重复性好.
