血清白蛋白和Dane颗粒的作用

HBeAg强阳性患者之血清,经密度区带离心和8ephadex G-200分离后,可以得到具有极高DNA一多聚酶活力的Dane颗粒.对提纯的Dane颗粒的研究表明,这种颗粒结合了一定量的白蛋白,结合的白蛋白在PAGE上和人血清白蛋白单体无明显差别,并非以多聚形式出现.对Dane颗粒和白蛋白结合状态的研究表明,这种结合是非共价的,PAGE可以使其分离.体外条件下,随着时间的推移,这种结合逐渐离解,具有可逆性,提示Dane颗粒和白蛋白的结合可能是以受体和配体的形式出现.目前普遍认为病毒可能通过白蛋白的桥联作用而入侵肝细胞,本文对血清白蛋白可能起的作用进行了讨论.%Purified Dane partioles were obtained from HBeAg positive serum by sucrose gradient ultraeentrifugation and sephadex G-200 gel filtration.It could be reaoted with anti-albumin antibody and a precipitating line was formed in counter immune-eleotrepharesis,but the reaotion could be inhibited by anti-HBs antibody.After stored at 4℃ for two months,another preoipitaring line was formed in same condition and could not be inhibited by anti-HBs antibody.However.the sample was further incubated at 37℃ overnight,the line disappeared.The binding albumin from Dane particle was proved as the monomer.type of serum albumin by-non-reducing PAGE.These results suggest that the binding between Dane particle and albumin is reversible.It is consistent with the hypothesis thab albumin is combined with viral particle in receptor and ligand form.It could explain the imporbant role of serum albumin as the link between HBV and hepatooytes.
