Foxp3在Graves病患者外周血单个核细胞中的表达及临床意义

         

摘要

目的:研究Graves病患者外周血叉头/翼状螺旋转录因子(Foxp3)的表达,探讨其在Graves病免疫发病机制中的作用.方法:取32例Graves病(GD)患者外周血单个核细胞(PBMC),经细胞表面及胞内双重染色后通过流式细胞术(FCM)测定Foxp3的表达,同时采用电化学发光免疫分析法(ECLIA)测定血清甲状腺激素、促甲状腺激素受体抗体(TRAb)的水平.结果:GD组治疗前外周血Foxp3阳性率显著低于正常对照组(P<0.01),且与同时测定的FT3、FT4呈显著负相关(P<0.01);治疗后,随着甲状腺功能的逐渐改善Foxp3水平虽仍低于正常对照(P<0.05)但较治疗前明显升高(P<0.05).结论:GD患者体内存在Foxp3的表达异常,Foxp3可能参与了GD的发病,而高甲状腺素血症与Foxp3调节性T细胞两者之间有一定关联.%Objective: To investigate the expression and significance of Foxp3 in Graves disease( GD) patients. Methods: The level of Foxp3 in peripheral blood mononuclear cells from the GD patients was evaluated by flow cytometry with surface and intracellular double staining in vitro,and the levels of serum thyroid hormone and TRAb were measured by electro-chemiluminescence immunoassay(ECLIA) .Results:Compared with that of the normal control group,the levels of Foxp3 in untreated GD patients were significantly lower( P <0.01).A significant negative correlation( P < 0.01 ) between Foxp3 levels and thyroid hormone levels was found in this group of patients. Following treatment, Foxp3 levels were significantly higher than that before treatment( P < 0.05), but lower than that in the normal controls( P < 0.05). Conclusion:The expression of Foxp3 is abnormal in GD. Foxp3 may play a potential role in the progression of GD onset,and the interaction of Foxp3 with high levels of thyroid hormone levels might be implicated in the immunopathology of GD.
