肺靶向汉防己甲素缓释微囊的小鼠体内分布

             

摘要

OBJECTIVE To determine the concentration of tetrandrine(TET) in biological samples, and study the distribution in mice for microencapsulated TET.METHODS An RP-HPLC TET was established changes in TET concentration was measured as a function of time after iv administration of TET injection or TET encapsulated albumin microcapsules. The parameters of targetability were also calculated.RESULTS The established RP-HPLC method is simple and accurate. Concentrations of TET in lungs for TET encapsulated albumin microcapsules dosed mouse via iv were higher than those in other organs,and decreased slowly.For TET injection group,concentrations of TET in livers were the highest. The microencapsulated TET had well lung-targeted characteristics.CONCLUSIONS The microencapsulation technique can deliver TET mainly to lungs, in which TET was released slowly from microcapsules.These were very important for the therapy of pulmonary hypertension in clinic.%目的:测定汉防己甲素在体内样品中的浓度,研究汉防己甲素经微囊化后的体内分布特点。方法:建立生物样品中汉防己甲素浓度测定的RP-HPLC法,对比汉防己甲素水针剂和微囊混悬液经小鼠尾静脉注射后,不同时间时汉防己甲素的浓度值,分析药物的肺靶向性。结果:生物样品检测方法简便易行,结果准确,微囊化汉防己甲素在肺中的浓度明显提高,且缓慢降低,具有明显的肺靶向性。结论:汉防己甲素经微囊化后,可浓集于肺部,且具有缓慢释放的特点,对肺动脉高压的防治具有重要意义
