血管内皮生长因子在非小细胞肺癌患者中的表达及其与临床病理参数的相关性

         

摘要

目的 检测血管内皮生长因子(VEGF)在非小细胞肺癌(NSCLC)患者组织和血清样本中的表达,探讨VEGF表达与NSCLC分期和肿瘤大小之间的相关性,以及VEGF对NSCLC细胞系增殖能力的影响.方法 对NSCLC组织和血清标本以及NSCLC细胞系A549和95C进行qRT-PCR、ELISA和MTT细胞增殖检测.收集30对NSCLC组织和相邻的非肿瘤肺组织标本以及10名健康志愿者的血清样品.结果 NSCLC组织和血清VEGF表达均明显上调,并且这种趋势和组织的临床阶段发展之间具有显著的正相关性.另外,外源性VEGF刺激明显促进了A549和95C细胞株的增殖能力,同时VEGF中和性抗体使A549和95C细胞株的增殖能力得到抑制.结论 VEGF能够促进NSCLC细胞增殖,其表达水平可能是NSCLC疾病发展中有用的生物标志物.%Objective To detect VEGF level in the non-small cell lung cancer (NSCLC) patients tissues and serum specimens and investigate the relationship between VEGF expression and tumor stage and tumor size of NSCLC as well as the influence on proliferation of NSCLC cell lines. Methods Quantitative Real Time Polymerase Chain Reaction (qRT-PCR), Enzyme Linked Immunosorbent Assay (ELISA), tumor cell proliferation by MTT were performed on human NSCLC tissues and serum specimens as well as on human NSCLC cell lines A549 and 95C. Thirty paired NSCLC tissue and adjacent non-tumor lung tissue specimens and ten cases serum samples of healthy volunteers were collected. Results Both tissue and serum VEGF expression were dramatically up-regulated in NSCLC patients as compared to their normal controls, furthermore, they were revealed to have a positive correlation with clinical stage development. In addition, extrinsic VEGF treatment remarkably promoted proliferation ability of A549 and 95C cell lines, while VEGF neutralizing antibodies inhibited the proliferation of A549 and 95C cell lines. Conclusion VEGF had ability to enhance the NSCLC cell proliferation and its level may be a promising biomarker for the development of NSCLC.
