ACS患者单核细胞MMP-9及其抑制物TIMP-1mRNA的表达

         

摘要

Objective To study the relationship between the expressions of MMP-9 and TIMP-1 mRNA in circulating monccytes and acute coronary syndrome (ACS). Methods By the same method, circulating monocytes were separated from 8 patients with ACS,8 patients with stable angina pectoris(SAP) and 8 persons without coronary heart disease (CHD). Then the total RNAs was isolated by acid guanidium thiocyanate-phenol-chloroform method from these cells and the expressions of MMP-9 and TIMP-1 mRNA were examined by RT-PCR. Furthermore the coronary stenesis was scored according coronary arteriography and the value of serum ox-LDL was detected. Results The expression of MMP-9 mRNA in monocytes of ACS patients was increased significanfiy compared to the patients with SAP and controls ( P < 0. 05). But there was not significant difference in expression of TIMP-1 mRNA. The serum level of ox-LDL in ACS patients was increased significantly compared to the patients with SAP and controls ( P <0. 05). Furthermore the senna level of ox-LDL in ASP patients was also increased significanfiy compared to cantrols( P < 0. 05 ). The expression of MMP-9 mRNA was correlated with serum ox-LDL level( r = 0. 458, P =0. 024), but not with the extent of atherosclerotic lesions. Conclusions The imbalance expressions of MMP-9 mRNA and TIMP-1 mRNA could be involved in the development and disruption of unstable plaque.%目的 探讨单核细胞基质金属蛋白酶-9(MMP-9)及组织型基质金属蛋白酶抑制物-1(TIMP-1)mRNA表达与急性冠脉综合征(ACS)之间的关系.方法 分离8例ACS、8例稳定性心绞痛(SAP)及8例对照循环血单核细胞,采用异硫氰酸胍-酚-氯仿抽提法提取总RNA,扩增目的 基因MMP-9及TIMP-1 mRNA.根据冠脉造影进行冠脉狭窄程度计分并测定血清ox-LDL水平.结果 ①ACS患者外周血单核细胞MMP-9 mRNA表达与对照组比较增加(P<0.05),而TIMP-1 mRNA表达在各组间无差异;②ACS组血清ox-LDL浓度高于SAP组及对照组(P<0.05),且SAP组高于对照组(P<0.05),③单核细胞MMP-9 mRNA表达与血清ox-LDL浓度正相关(r=0.458,P=0.024);而与冠脉狭窄计分(CSS)无相关性.结论 ACS患者外周血单核细胞MMP-9、TIMP-1 mRNA表达比例失衡,可能直接参与了不稳定斑块的形成及破裂.
