目的 观察黄连解毒汤(HLJDT)对代谢综合征(MS)大鼠心肌的保护作用及对胰岛素信号转导的影响,探讨其保护心肌的分子机制.方法 高糖、高脂及高盐饲料及20%高蔗糖水喂养(HF饮食)喂养Wistar大鼠建立MS大鼠模型,以HLJDT干预治疗12 w,检测血糖、血清胰岛素和血脂水平,Western印迹方法检测HLJDT治疗后MS大鼠心肌组织内核因子-kB(NF-kB)、磷酸化Akt(P-Akt)蛋白表达及胰岛素受体底物1(IRS-1)蛋白色氨酸磷酸化水平.结果 HLJDT治疗MS大鼠后,心肌组织内NF-kB表达及IRS-1丝氨酸磷酸化水平较MS组下降,P-AKT显著增加.结论 黄连解毒汤通过抑制炎症降低IRS-1丝氨酸磷酸化水平,这可能是其保护心肌的机制之一.%Objective To investigate the effects of Huanglian Jiedu decoction (HLJDD) on myocardium and signal transduction of insulin receptor substrate in rats of metabolic syndrome ( MS) , and explore its possible molecular mechanism of improving myocardial lesions. Methods MS rats were established with obese-diets (a high-fat, high-glucose and high-salt diet and sucrose solution). MS rats in the testing group were treated with HLJDD for 12 weeks. Fasting serum glucose, insulin and blood-fat were determined, and Western blot was performed to analyze the proteins in signal transduction pathway, including nuclear factor kappa B (NF-kB) , phosphorylation Akt and tyrosine phosphorylation levels of insulin receptor substrate-1 (IRS-1). Results NF-kB protein and tyrosine phosphorylation and IRS-1 were decreased, and phospho-AKT protein was increased significantly in MS rats treated with HLJDD, compared with untreated MS rats. Conclusions HLJDD could protect myocardium from IR-mediated injury by inhibiting IRS serine phosphorylation catalyzed by inflammation in MS rats.
