蒙药嘎日迪-13味丸对脑缺血大鼠海马组织白介素-1β、肿瘤坏死因子-α蛋白及神经细胞凋亡的影响

         

摘要

目的:探讨蒙药嘎日迪-13味丸对脑缺血大鼠海马组织炎性细胞因子白介素-1β( IL-1β)和肿瘤坏死因子-α( TNF-α)表达及神经细胞凋亡的影响。方法采用枕大池二次注血法至大鼠脑组织缺血损伤模型,在各时间段断头取脑,用 ELISA 法和流式细胞仪分别检测脑缺血海马组织中 IL-1β和 TNF-α的含量及细胞凋亡数量的百分比。结果与正常组比较,模型组海马组织中 IL-1β和 TNF-α升高,细胞凋亡百分比增多( P<0.05);与模型组比较,嘎日迪-13味丸组大鼠海马组织中 IL-1β和 TNF-α含量降低,细胞凋亡百分比数减少( P<0.05)。结论嘎日迪-13味丸能降低大鼠脑缺血组织神经细胞凋亡数量,减轻缺血损伤程度,其机制可能与降低脑组织内 IL-1β和 TNF-α含量有关。%Objective To investigate the effects of Mongolian medicine Garidi -13 pill on inflammatory cytokines interleukin-1β( IL-β) and tumor necrosis factor-α(TNF-α) expression and nerve cell apoptosis of cerebral ischemia hippocampus tissue of rats .Methods Two times of blood injection to the cistema magna was used to make the rat model of cerebral ischemia .Brain tissue was acquired at each time point.Flow cytometry and ELISA were used to detect the number of apoptotic cells , IL-1βand TNF-αcontent in hippocampus tissue.Re-sults Compared with normal group, IL-1βand TNF-αin hippocampus were increased , apoptosis percentage number was increased of model group (P<0.05).Conclusions Garidi-13 can reduce the number of apoptosis nerve cell in ischemia brain tissue and reduce the severity of ischemic injury, its mechanism may be related to reduce IL -1βand TNF-αcontent of brain.
